冻干口服霍乱rBS－WC菌苗安全性及免疫原性的人群试验

摘　　要：为考察国产冻干重组霍乱毒素Ｂ亚单位－灭活霍乱弧菌全菌体（ｒＢＳ－ＷＣ）口服菌苗的安全性及免疫原性，将３６９人随机分为３组，采用双盲法分别服用高剂量菌苗（５ｍｇｒＢＳ＋１×１０＂ＷＣ）、低剂量菌苗（１ｍｇｒＢＳ＋１×１０＂ＷＣ）及安慰剂。服苗方案为０、７、２１天。试验结果显示，服苗组的不良反应率为０．４％（１／２４７），对照组无不良反应；免疫组血清抗体和粪抗体的阳转率及抗体水平均显著升高；两免疫组间血清及粪抗体阳转率及抗体水平差异无显著性；对不同性别、年龄及服苗次数，抗体的产生有所不同。结果表明。Inordertoevaluatethesafetyandimmunogenicityofdomesticproducedlyophilizedrecombinant,B-subunit,inactivatedwholecelvaccineofvibriocholerae(rBS-WC),369subjectswererandomlydividedintothreegroupsandobservedwithmaskingmethod,onewithhighdoseofvaccine(5mgrBSand10'WC),anotherwithlowdose(1mgrBSand10'WC),andthecontrolonewithplacebo.Threedosesofvaccineweregivenoralyatanintervalofsevendaysand14days,re-spectively.Resultsshowedthatonlyonesubjecthadmildadversereactioninthevaccinegroup(1/247)andnoneinthecontrolgroup.Serumandfecalantibodyconversionratesandaveragelevelsofantibodiesinthetwovaccinegroupsweresignificantlyhigherthanthoseincontrolone.Therewasnosignificantdiferenceinserumandfecalantibodyconversionrateandaverageantibodylevelsbe-tweenthetwovaccinegroups.But,antibodyresponsewasdifferentamongsubjectsofvarioussex,age,andvaccinedoses.ItindicatesthatrBS-WCvaccineofvibriocholeraehadgoodimmunogenicityandsafety.
