摘  要:目的:建立雌激素缺乏的动物模型并在此基础上建立腹主动脉瘤(abdominalaorticaneurysm,AAA)模型,研究雌激素缺乏对AAA形成中的影响及其作用机制。方法:雌性Wistar大鼠20只,随机分为实验组和对照组,各10只。实验组(切除卵巢)和对照组(假手术)分别于术后1周用弹性蛋白酶灌注肾下腹主动脉制成AAA模型。14d后用放射免疫法检测血清E2的含量,用免疫组化及RealTimeRTPCR测定动脉瘤组织基质金属蛋白酶(matrixmetalloproteinase,MMP)2和MMP9的mRNA及蛋白的表达。结果:实验组与对照组比较雌激素水平显著降低,P<0.01。其动脉瘤扩张率分别为(160.52±18.38)%和(101.30±6.73)%,P<0.01;MMP2mRNA的相对表达量分别为0.128±0.070和0.014±0.006,P<0.01;MMP9mRNA的相对表达量分别为1.50±0.43和0.33±0.11,P<0.01;MMP2的表达率分别为(48.51±9.63)%和(25.33±3.15)%,P<0.01;MMP9的表达率分别为(46.46±5.91)%和(24.78±2.30)%,P<0.01。结论:在AAA形成的过程中雌激素缺乏可导致MMP2和MMP9的mRNA表达增强,MMP2和MMP9合成及炎性细胞浸润的增加,进一步促进AAA的形成。OBJECTIVE: To investigate the mechanism of estrogen deficiency in the formation and development of experimental abdominal aortic aneurysm. METHODS: Female Wister rats were divided into experiment group and control group randomly. Rats in experiment group were ovariectomized and those of control group were sham-operated. Elastase perfusion of the infrarenal aorta was performed in rats. At 14 days, estrogen was detected by radioimmunoassay and aortas were harvested for immunohistochemistry and real-time RT-PCR. RESULTS: Experimental group showed a lower estrogen level than that of control group, P〈0.01. Aneurysms dilatation rates of experiment group and control group Were(160.52±18.38)% and (101.30±6.73)% respectively(P〈0.01). Their relative expression quantities of MMP-2 mRNA were 0. 128±0.070 and 0.014±0.006 respectively(P〈0.01) and those of MMP-9 mRNA were 1. 50±0.43 and 0.33±0. 11 respectively(P〈0.01). MMP-2 expression of experiment group and control group were (48. 51±9.63) and (25.33±3.15)% respectively (P〈0.01) and those of MMP-9 were (46.46±5.91)% and (24.78±2.30)% respectively (P〈0.01). CONCLUSIONS: In AAA development, estrogen deficiency may promote the AAA formation by enhance MMP-2 and MMP-9's expression of mRNA and protein and inflammatory cell infiltrate.
