摘 要:目的:研究大鼠供体喉的保存,探讨鼠喉保存的评价方法及保存时间。方法:采用Wister大白鼠,切取供体喉后用UW氏液进行灌洗,并低温保存至不同时段,分别通过病理光学及透射电镜观察其形态学改变。结果:病理光镜检查在移植喉保存60 h后,才能发现大鼠喉组织的缺血性改变,透射电镜可以在鼠喉保存24 h后发现细胞的超微结构改变,28 h后供体喉大部分组织出现细胞核固缩和凋亡。结论:透射电镜能及早地反映喉体组织缺血和坏死情况,优于病理光镜检查,UW氏液保存移植喉可达26 h。OBJECTIVE:To study the preservation of rat donor laryngeal allograft evaluation methods and preservation time limit larynx and discuss. METHODS: After harvesting laryngeal allograft of Wister rat, it was perfused by UW's solution and preserved under cold condition at different intervals. The continue changes of donor rat larynx were observed under light microscopy and electron mi- croscopy were perforned. RESULTS:The ischemic change was found by HE stain at 60 hours. Ultrastructure of laryngeal allograft cells appeared ischemic injury at 24 hours under electron microscopy. Most tissues of allograft appeared pyknosis and apoptosis 28 hours later. CONCLUSIONS: Electron microscopy can detect ischemic or necrosis of laryngeal allograft earlier and better than light microscopy. UW solution can preserve larynx for 26 hours.
