摘  要:目的 以盐酸哌唑嗪为模型药物研究单层芯渗透泵片简化的制备方法。方法 用自行设计的带针冲头压制带孔洞的片心.然后以乙基纤维素为包衣材料包衣得单层芯渗透泵片。研究片心处方、孔径等因素对渗透泵片释药行为的影响。结果 将片心处方实验结果进行多元回归和优化处理得到了优化片心处方。自制渗透泵片在24h内释药速率平稳。在0.80~1.15mm内,片心上孔径对自制渗透泵片的释药行为影响不大。结论 用带孔片心包衣制备渗透泵片的方法免去了渗透泵片制备中的激光打孔机,大大简化了渗透泵片的制备工艺。OBJECTIVE To develop a simplified preparative method for osmotic pump tablet using prazosin hydrochloride as a model drug.METHODS The core tablets with an indentation were compressed by the punch with a needle. The osmotic pump tablet was prepared by coating the indented tablet with ethyl cellulose. The influences of core formulation variables and the indentation size of the core tablet on drug release were investigated. RESULTS A multiple regression equation was obtained with the experimental data of core tablet, and then the formulation was optimized. The optimal osmotic pump tablet was found to be able to deliver prazosin hydrochloride at a rate of approximate zeroorder up to 24 h. The orifice size of the core tablet hardly affected drug release in the range of (0.80 - 1.15) mm. CONCLUSION The osmotic pump tablet is made from the core tablet with an indentation. This method exempts the laser drilling and makes the preparation of osmotic pump tablet very simply.
