清利固精颗粒对系膜增殖性肾炎肾组织PDGFmRNA表达的影响

摘　　要：目的：探讨清利固精颗粒对大鼠抗Thy-1系膜增殖性肾炎（MsPGN）肾组织PDGF-A、BmRNA表达的影响。方法：本研究应用大鼠抗Thy-1系膜增殖性肾炎（MsPGN）动物模型，将大鼠随机分为4组：正常对照组、模型组、肾炎康复片组、清利固精组，每组8只大鼠。各实验组于第7天处死大鼠，采用RT-PCR检测肾组织PDGF-A、BmRNA表达含量。结果：模型组大鼠肾组织PDGP-A、BmRNA表达比正常对照组明显升高（P〈0．05～0．01）；与模型组相比，清利固精组PDGF-A、BmRNA表达明显下降（P〈0．05～0．01）；清利固精颗粒降低肾组织PDGF-BmRNA表达疗效优于肾炎康复片（P〈0．05）。结论：清利固精颗粒剂抑制细胞增殖及细胞外基质增加，可能与其能降低MsPGN肾组织PDGF-A、BmRNA表达有关。Objective： To investigate the effect of QGG on the expression of PDGF - AmRNA and PDGF - BmRNA in rat kidney tissue with MSPGN. The model was induced by reped injedction of anti - Thyl antiserum. Methods： 32 rats were randomly divided into four groups, the control group, the MSPGN group, the Shenyan Kangfu Tablet（SKT） - treated group and the QGG - treated group. Rats were taken on day 7 to measure on the expression of PDGF - A mRNA and PDGF - BmRNA in rat kidney tissue with RT - PCR. Results ： On the expression of PDGF - A mRNA and PDGF - BmRNA in rat kidney tissue in the MSP（;N rats as compared with the control rats（ P 〈 0.05 - 0.01 ）. The expression of PDGF - A mRNA and PDGF- BmRNA in rat kidney tissue decrease in the QGG - treated rats compared with the MSPGN rats （P 〈 0.05 - 0.01 ）. And the effect of decreasing the expression of PDGF - BmRNA in the QGG - treated rats was better than the SKT - treated rats （ P 〈 0. 05）. Conclusion： QGG can inhibit mesangial cell and extra cellullarmatrix proliferation. The mechanism may be it can decreae the expression of PDGF - AmRNA and PDGF - BmRNA in kidney tissue of the MSPGN rats.
