反相高效液相色谱法同时测定保健口服液中20种甾体激素的研究

摘　　要：建立了利用反相高效液相色谱法同时测定保健口服液中２０种甾体激素的方法，进行了色谱条件的优化和样品预处理方法的探讨，并应用于市售保健口服液中微量激素的测定。最低检出量为０．１６～３．１８ｎｇ，回收率为８０．５％～１０３．１％，相对标准偏差为３．４％～９．２％。A method for determination of 20 kinds of steroid hormones in health-caring drinkings by revered phase high performance liquid chromatography has been established. The steroid hormones to be analyzed include estrogens, androgens, progestogens and corticosteroids. The chromatographic conditions were as follows:PE C18 column, 150mm × 4. 6mm, at 40℃, gradient elution with methanol-20mmol/L phosphate buffer (pH 7. 0) system and double wavelength detection at 245and 280nm. Zearalenone was selected as the internal standard. The hormones in the health-caring drinkings were extracted with CHCl3-CH3CN (9/1) and washed thoroughly with water so as to eliminate the water-soluable impurity peak in chromatogram. By means of this method, the trace hormones in the health-caring drinkings from market have been determined. At the same time, these samples were also analyzed by radioimmunoassay. The results of two methods coincide very well.The superiority of this method is its high extraction efficiency and less interference. So this method is simple,fast, inexpensive and suitable for wide applicaion. The minimum detectable quantities of steroid hormones in health-caring drinkings were in the range from 0. 16ng to 3. 18ng. The recoveries ranged from 80. 5% to 103. 1 % with RSD from 3. 4% to 9. 2%.
