人生长激素抗独特型抗体的制备及免疫学鉴定

摘　　要：目的制备人生长激素抗独特型抗体,并进行鉴定。方法应用人生长激素抗原(Ag)纯品,通过杂交瘤技术制备人生长激素单克隆抗体(Ab1),经免疫亲和层析柱纯化后,免疫家兔,其血清经饱和硫酸铵纯化后,获得人生长激素抗独特型抗体(Ab2)。再应用ELISA法进行鉴定。结果Ab2与Ab1呈阳性反应,与BALB/c小鼠γ球蛋白呈阴性反应。Ab2与Ag竞争结合Ab1,Ab2抑制Ab1与Ag的结合。结论已成功制备针对人生长激素的单克隆抗体和具有人生长激素抗原内影像的抗独特型抗体。Objective To prepare and identify the anti-idiotype antibody against human growth hormone. Methods Prepare monolclonal antibody Ab1 with the antigen（Ag） of human growth hormone by hybridoma technique and purify by immunoaffinity chromatography. Immunize rabbits with Ab1 and purify their sera by saturated ammonium sulfate precipitation to obtain anti-idiotype antibody Ab2 against human growth hormone. Identify Ab2 by ELISA. Results Ab2 showed positive reaction with Ab1 but no reaction with γ-immunoglobulin of BALB/c mice. Ab2 competitively inhibited the binding of Ab1 to Ag. Condusion The monoclonal antibody against human growth hormone and anti-idiotype antibody with the internal image of antigen of human growth hormone were successfully prepared.
