摘 要:本文对成都大熊猫繁育研究基地、成都动物园、福州大熊猫研究中心及昆明动物园圈养繁殖的大熊猫14胎次、19只幼子、25只次参配雄兽个休的被毛、血液、精液、肝、甲醛固定组织细胞,以及粪、尿等样品,采用同位素与荧光素标记基因探针作DNA指纹图谱分析,得到了一些有意义的结论:(1)同一大熊猫个体的被毛、血液、精液、肝、甲醛固定组织细胞及粪、尿检测,其DNA指纹图完成一致。(2)大熊猫一雄配一雌.或多雄配一雌,受配雌善所产任一个子代个体的DNA指纹条带,均分别来自父亲、母亲.以及父母双亲,没有新带出现,无误的确定了19只子代的真正父亲.排除了非父。(3)5个双胞胎,即10只子代个体的DNA指纹图谱,均具个体特异性,表明为异卵双生,其中4个双生子个体为异卵异父双生,为大熊猫在一个发情期不一定同时排卵提供了佐证,亦为多雄配一雌,以及多次配种提供了理论和应用依据。(4)DNA指纹技术用于大熊猫的父权认定,灵敏稳定.准确无误。(5)应用寡核苷醚基因探针LZF—l作大熊猫亲子鉴定较JH12.6探针更为适合。(6)荧光素及同位素标记探针LZF—l其效果基本一致。因此荧光素标记可以取代同位素32P标记方法。he paternity determination of filial generation of artificial breeding Giant Panda including 19young babies and 25 'father' in 14 litters in Chengdu Research Base of Giant Panda Breeding,Chengdu Zoo,Fuzhou Giant Panda Research Centre and Kunming Zoo has been done by using the method of labelling oligonucleotide probe LZF-1 and JH 12. 6 with fluorescein and radioactive P,and the DNA were isolated from hairs, blood, semina, formalin-fixed testes,excrements and urine. (1 )There are the identical results of DNA fingerprints by detecting the hairs, blood,semina,formalin-fixed testes,excrements and urine of the same Giant Panda. (2)All the DNA hybrid bands of any young baby farrowed down by monogamy and polyandry including 2,3 and 4 males can been observed in their parents,and the true fathers of the 19 young babies have been determined. (3)The DNA fingerprints of 10 young babies in 5 pairs of twins are highly individual spe cific,and it has been known that the 5 pairs of twins are the freternal twins. In these fraternal twins 4 babiesborn in 1995 in 2 pairs of twins in Chengdu Research Base of Giant Panda Breeding and Chengdu Zoo have their respective fathers,and it has made showed that the filial generation of artificial breeding Giant Pandas havenotonly the fraternal twins but also the 'twins of half blood'. The results have provided the theoretical and applied basis for raising farrowing efficiency of female Giant Pandas by polyandry, mating or inseminating many times in an oestrus. (4 )The method of detecting the DNA fingerprints in the paternity determination of Ginat Pandas is sensitive and stable,it has solved the fear of disturbance in the rear for the mating ways of polyandry, mating or inseminating many times in an oestrus of Giant Panda. (5 ) The detecting results of oligonucleotide probe LZF-1 are better than that of JH 12. 6,and the method of labelling LZF--1 with fluorescein can totally replace that of labelling probe with radioactive P.
