摘  要:目的探讨AGM区基质细胞对异基因骨髓移植后造血重建及GVHD的影响。方法以7.5Gy60Co γ射线照射的BALB/ C小鼠为骨髓移植模型,将小鼠分为四组,A:正常对照组、B:空白对照组、C:异基因骨髓单个核细胞单纯移植组、 D:异基因AGM区基质细胞和骨髓单个核细胞联合移植组,计数并比较各组小鼠骨髓移植后第4、11、18d的WBC及BMMNC 数目,观察并记录比较C、D两组小鼠的一般情况和存活时间及存活率等各项GVHD指标。结果联合移植组在照射后第 11、18d的白细胞及骨髓单个核细胞计数均高于单纯移植组,至第18d已趋近于正常。且联合移植组GVHD发生时间较晚, 存活率显著高于单纯移植组。结论 AGM区基质细胞不仅能加快造血重建,还能延缓移植后GVHD的发生,降低死亡率, 提高移植成功率。Objective Discuss the influence of stromal cells in AGM region (AGM-S) on hematopoietic recon- struction and acute graft versus host disease (GVHD) after ALLo-BMT. Method The BALB/C mice radiated by 7. 5y ^60Co gamma rays are taken for BMT model. These mice are divided into four groups as follows:A:normal control group;B:blank control group;C: BMT alone group of allogene BMNNC;D: cotransplantation group of allogens AGM- S and BMNNC. The WBC and BMMNC number of mice in each group 4,11 and 18 days after BMT are counted and compared. The GVHD targets of group C and D, such as general situation,survival time and survival rates,are observed and compared. Result The WBC and BMMNC cell counts of cotransplantation group are significantly higher than those in the group with BMT alone 11 and 18 days after irradiation,and are close to normal group in the 18th day. The time of GVHD of cotransplantation group is later than that of BMT alone group,amd the survival rate is much higher.Conclusion AGM-S not only can promote the hematopoietic reconstruction,but also can postpone the GVHD after BMT, lower the death rate and enhance the success ratio of BMT.
