摘 要:目的探讨穴位区细胞外基质在针刺信号转导中的作用机制。方法SD大鼠40只,雌雄各半,随机分为4组,每组10只,以大鼠热辐射甩尾实验的温度变化为观察指标。实验组大鼠“后三里”区注射人工合成的五肽甘氨酰-精氨酰-甘氨酰-天冬酰-酪氨酸GRGDY(G ly-Arg-G ly-Asp-Tyr)30 m in后再针刺,以大鼠基础痛阈组、“后三里”区注射0.9%NaC l组及针刺后三里组为对照。结果GRGDY注射后30 m in再针刺,大鼠甩尾的温度与针刺组相比未见明显改变。结论GRGDY未能阻断针刺效应,针刺效应的初始机制可能与细胞外基质与整合素特异性的结合位点RGD无关。Projective To investigate the mechanism of action of point-regional extracellular matrix in acupuncture signal transmission. Methods Forty SD rats, half male and half female, were randomly allocated to four groups, 10 each. A change in temperature in a rat heat radiation tail wagging test was taken as an observation index, The experiment group of rats was acupunctured 30 min after an injection of "Housanli" area with synthetic pentapeptide GRGDY: glycyl-arginyl-glycyl-aspartyl-tyrosine (Gly-Arg-Aly-Asp-Tyr). The basic pain threshold group of rats, the group of rats in which "Housanli" area was injected with 0.9% normal saline and the group of rats in which Housanli was acupunctured were taken as controls. Results The temperature as which the rat wagged its tail did not significantly change after GRGDY injection and acupuncture following 30 min later as compared with that in the acupuncture group. Conclusion GRGDY can not prevent the effect of acupuncture. The initial mechanism of acupuncture effect may be not related to extracellular matrix RGD combining with integrin specifically.
