摘  要:目的通过对T淋巴细胞亚群的检测,从免疫学方面探讨同种异体骨髓基质细胞组织工程化骨植入兔体内修复长段骨缺损的可行性。方法用骨髓基质细胞(MSCs)、重组人骨形态蛋白2(rhBMP2)与纤维蛋白胶(FG)复合培养,体外构建组织工程化骨,植入5只异体兔的桡骨1.5cm节段骨缺损作为实验组;另5只兔用单纯FG植入同样大小骨缺损作为对照组;分别于术后1、2、3、6、8周抽取静脉血,应用流式细胞术检测样本中T淋巴细胞亚群CD4+、CD8+阳性率并计算CD4+/CD8+比值。结果术后实验组和对照组的1、2、3、6、8周各时间点T淋巴细胞亚群CD4+、CD8+和CD4+/CD8+比值分别与术前的T淋巴细胞亚群两两相比,两者阳性率差异均无显著性(P>0.05)。结论FG、rh-BMP2和兔MSCs复合构建组织工程化骨植入异体兔体内其术后8周内免疫反应水平低,可用以修复兔节段骨缺损。Objective To investigate the effects of tissue engineered bones on T lymphocyte subsets after bridging segmental bone defects in rabbits. Methods MSCs aspirated from rattits'greater troochanter of femur were cultured in vitro and amplified. Then the MSCs, rhBMP2 and Fibrinogen were made into a complex and implanted into 5 allogenic rabbits to bridge 1.5cm segmental bone defects as experiment group. Fibrinogen was implanted to bridge same size defects in other 5 allogenic rabbits as control group. The CD4^+ , CD8^+ , CD4^+/CD8^+ T lymphocytes in blood were detected using flow cytometer at 1,2,3,6 and 8 weeks after the operation. Results T lymphocyte subsets CD4^+ , CD8^+ and CD4^+/CD8^+ at every time point after the operation in the experiment group and control group had no significant difference as compared to pre-operation. Conclusion Tissue engineered bones constructed with Fibrinogen, and rhBMP 2 and allogenic MSCs showed low immunogenicity within 8 weeks and could be implanted to repair bone defects in rabbits.
