摘 要:作者在建立检测庚型肝炎病毒酶免疫和PCR技术的基础上,将抗-HGV阳性病人血清经逆转录─巢式PCR分离部分HGVcDNA片段,用与HGV基因互补的特异寡酸引物进行双脱氧核苷酸链末端终止法──PCR直接测序。结果表明,中国大陆HGV-302Ⅰ株部分cDNA序列与Linnen等报道的HGV美国株cD-NA序列有极高的同源性。5’端非编码区、前1区、包膜E1区、NS3区和NS5区与美国株cDNA序列有高达90%以上的同源性,编码相应氨基酸的同源性达96%。表明HGV感染虽呈全球分布,但在进化上属遗传高度保守。The partial cDNA genes of hepatitis G virus(HGV) were isolated from serum of a patient with chronic hepatitis C virus infection.Both of the positive anti HGV and the HGV RNA were detected by ELISA and PCR.The partial cDNA fragments from the 5′ non coding region、Pre E1、NS3 and NS5 regions were amplified.The total 655 nt of these cDNA fragments were sequenced and analysed.The results showed that the partial cDNA sequences from this patient was 92%~95% nucleotide identity and over 96% amino acid identity with the sequences of HGV cDNA in USA which was reported by Linnen et al .in 1995.It was shown that the HGV infection has a global distribution and the greatly homology of gene sequences of HGV in different countries will be beneficial to the development of universal vaccine and dignosis reagents.
