解毒化瘀Ⅱ方对肝衰竭大鼠肝线粒体内Ca^(2+)稳态的影响

摘　　要：目的:观察解毒化瘀Ⅱ方对暴发性肝衰竭大鼠肝线粒体内Ca2+稳态的影响。方法:实验于2004-05/2005-12在广西中医学院中医肝病治疗中心实验室完成。选择Wistar大鼠84只,随机数字表法分为空白组、模型组、解毒化瘀Ⅱ方低、中、高剂量组、乳果糖组、安宫牛黄丸组,每组12只。解毒化瘀Ⅱ方由茵陈30g,赤芍50g,白花蛇舌草30g,大黄15g(后下),郁金15g,石菖蒲15g组成,低、中、高剂量组分别给予生药9.1,28.76,57.55g/kg、乳果糖组给予3.5g/kg、安宫牛黄丸组给予1.5g/kg。使用时,用蒸馏水将解毒化瘀Ⅱ方的配方颗粒剂配成0.033,0.11,0.22g/mL,乳果糖配成0.0875g/mL,安宫牛黄丸配成0.0375g/mL,空白组与模型组分别给予蒸馏水代替,每天灌胃量40mL/kg。造模前3d开始灌胃给药,2次/d,间隔12h,共给药5d12h。除空白组外均采用皮下注射硫代乙酰胺法复制暴发性肝衰竭大鼠模型,造模完成12h后测定各组大鼠血清单胺氧化酶、谷草转氨酶、谷丙转氨酶、脱苷脱氨酶活性及肝线粒体内单胺氧化酶、钙镁三磷酸腺苷酶活性及Ca2+含量。结果:纳入动物84只,均进入结果分析。①模型组大鼠血清单胺氧化酶、谷草转氨酶、谷丙转氨酶、脱苷脱氨酶活性与空白组比较显著升高,差异有显著性意义(P<0.01);解毒化瘀Ⅱ方各剂量组能降低大鼠血清单胺氧化酶、谷草转氨酶、谷丙转氨酶、脱苷脱氨酶水平,并呈现量效关系,以解毒化瘀Ⅱ方高剂量组的作用效果最佳,其与模型组、解毒化瘀Ⅱ方低剂量组、乳果糖组、安宫牛黄丸组比较差异有显著性意义[分别为(377.08±69.01),(792.32±155.36),(657.96±141.70),(457.92±85.35),(445.42±78.52)μkat/L;(10.82±1.88),(22.13±5.36),(17.12±4.37),(13.36±3.36),(16.39±3.21)μkat/L;(10.29±1.63),(26.23±6.49),(18.08±1.72),(12.39±2.74),(14.70±3.51)μkat/L;(414.58±86.18),(943.19±219.88),(669.13±189.37),(507.10±105.85),(582.78±111.36)μkat/L,P<0.01]。②�AIM： To observe the influence of the detoxification and dissipation blood stasis （DDBS） formula Ⅱ on hepatic mitoehondrial Ca^2＋ homeostasis in rats with break-out hepatic failure. METHODS： The experiment was performed at the Laboratory of Liver Disease Treatment Center, Guangxi Traditional Chinese Medical University between May 2004 and December 2005. A total of 84 Wistar rats were arranged into seven groups with 12 in each by random grouping： blank group, model group, the lactulose treating group, the ＂Bezoare pill for resurrection＂ treating group, and three groups treated with DDBS formula Ⅱ ： low dose group, middle dose group, high dose group. The DDBS formula was composed of 30 g virgate wormwood herb, 50 g red peony root, 30 g hedyotic diffusa, 15 g rhubarb （decocted later）, 15 g terra merita and 15 g grassleaf sweetflag rhizome. The low, middle, high groups were administrated with doses of 9.1, 28.76, 57.55 g/kg respectively, while the lactulose group received 3.5 g/kg lactulose and the ＂Bezoare pill for resurrection＂ group received 1.5 g/kg. In administration, we applied the mixtures of distilled water with these drugs except the blank group and model group were with distilled water only, and the concentrates of the three DDBS formula Ⅱ groups were 0.033, 0.11, 0.22 g/mL respectively, that of lactulose group was 0.087 5 g/mL and that of ＂Bezoare pill for resurrection＂ group was 0.037 5 g/mL And the amount of gastric perfusion was 40 mL/kg daily. Administration of gastric perfusion was executed 3 days before model-establishing, twice a day with an interval of 12 hours, 5 days and 12 hours in total. All rats in these groups except those in the blank group were made into models of fulminant hepatic failure by hypodermic injection with thioacetamide （TAA）.
