摘 要:目的:观察逆灸关元穴对更年期大鼠促性激素释放激素、卵泡刺激素和黄体生成素等相关激素的影响,以及逆灸对更年期下丘脑-垂体-卵巢轴稳态维护的机制。方法:实验于2003-11/2004-06在北京中医药大学针灸学院针灸机理实验室完成。①选取鼠龄4个月的健康大鼠10只为年轻正常组,另将鼠龄10个月的大鼠70只数字表法随机分为2组,逆灸组(n=30),自然对照组(n=40),根据处死时的鼠龄,逆灸组又分12,14,16月龄3个时间点,自然对照组分10,12,14,16月龄4个时间点,每个时间点10只。②逆灸组于10月龄时采用艾炷直接灸大鼠关元穴,每次1壮,2次/周,灸后局部皮肤略潮红,共灸8周;其他2组不干预。③各组于相应时间点分批取材,采用放射免疫方法检测血浆卵泡刺激素、黄体生成素水平,下丘脑及血液促性激素释放激素水平。结果:大鼠80只全部进入结果分析。①血浆卵泡刺激素水平:自然对照组16月龄大鼠高于年轻正常组[(23.757±6.212),(10.565±0.817)U/L,P<0.05],逆灸组16月龄大鼠低于同月龄自然对照组[(17.952±2.231)U/L,P<0.05]。②血浆黄体生成素水平各组比较差异不显著(P>0.05)。③卵泡刺激素/黄体生成素比值:自然对照组16月龄大鼠大于年轻正常组(0.171±0.021,0.094±0.006,P<0.05),逆灸组16月龄大鼠小于同月龄自然对照组(0.125±0.007,P<0.05)。④下丘脑促性激素释放激素水平:自然对照组14,16月龄大鼠低于年轻正常组[(3.566±2.387),(3.032±0.602),(12.041±4.815)mg/L,P<0.05,0.01],逆灸组12,14,16月龄大鼠均高于同月龄自然对照组(P<0.05)。⑤血浆促性激素释放激素水平:自然对照组16月龄大鼠高于年轻正常组[(99.425±21.657),(23.816±6.058)ng/L,P<0.05],逆灸组12,16月龄大鼠显著高于同月龄自然对照组(P<0.05)。结论:更年期大鼠随月龄的增加,下丘脑-垂体-卵巢轴促性激素释放激素、卵泡刺激素和黄体生成素等相关激素逐渐紊乱,逆灸关元AIM:To analyze the mechanism of maintaining the order of hypothalamic-pituitary-ovarian (HPO) with preventive moxibustion by observing the effect on gonadotroph in hormone-releasing hormone (GnRH), follicle-stimulating hormone (FSH), luteinizing hormone (LH) of menopausal rats with preventive moxibustion on the Guanyuan point. METHODS:The experiment was conducted in the Laboratory of Acupuncture and Moxibustion College, Beijing University of Traditional Chinese Medicine between November 2003 and June 2004. ①Ten healthy rats of 4 months old were selected to be the normal young group, and another 70 rats of 10 months old were randomly divided into two groups: preventive moxibustion group (n=30) and natural control group (n=40). The preventive moxibustion group was divided into 12-month, 14-month and 16-month groups according to the age of rats, and the natural control group was divided into 10-month, 12-month, 14-month and 16-month groups with 10 rats in each subgroup. ② In the preventive moxibustion group, moxibustion was applied to rat's Guanyuan point directly at the 10^th month with one zhuang every time and twice a week for continuous eight weeks, and the local skin slightly turned red after moxibustion. The other two groups received no intervention.③Materials were drawn from each group at corresponding time-point in batch. Radioimmunity was used to detect the levels of GnRH, FSH and LH of menopausal rats with preventive moxibustion. RESULTS:A total of 80 rats were involved in the analysis of results.① Level of FSH in serum: it was higher in the 16-month natural control group than in normal young group [(23.757±6.212), (10.565±0.817) U/L,P 〈 0.05], and it was lower in 16-month preventive moxibustion group than the 16-month natural control group [(17.952±2.231)U/L,P 〈 0.05]② There were no significant differences in level of serum LH among each group (P 〈 0.05). ③The ratio of FSH and LH: it was greater in the 16-month natural control
