摘 要:目的:总述转化生长因子β/Smad信号传导通路研究相关进展及中药干预肾间质纤维化的可行性和优势。资料来源:检索Medline1996-01/2006-06与转化生长因子β/Smad信号传导通路与中药抗肾间质纤维化相关的文献,检索词“TGF-β/Smadsignaling,traditionalChinesemedicine,renalinterstitialfibrosis”,并限定文章语言种类为英语。同时检索中国期刊全文数据库2001-01/2006-06与转化生长因子β信号传导通路与中药抗肾间质纤维化相关的文章,限定文章语言种类为中文,检索词“转化生长因子β/Smad信号传导通路,中药,肾间质纤维化”。资料选择:对资料进行初审,选择与转化生长因子β/Smad信号传导通路与中药抗肾间质纤维化相关的文献。排除重复性研究。资料提炼:共收集到54篇关于转化生长因子β/Smad信号传导通路与中药抗肾间质纤维化的文献,13个实验或临床研究符合纳入标准。排除的41篇为重复性研究。资料综合:转化生长因子β/Smad信号传导通路可能是肾间质纤维化的最后的共同途径,干预转化生长因子β/Smad信号传导通路对阻断肾间质纤维化有着积极的意义。中药复方具有多组分的物质基础,理论上具有多途径与多靶点的作用特点,针对转化生长因子β/Smad信号传导的复杂网络可发挥综合作用优势。结论:以转化生长因子β/Smad信号转导途径为基础,研究有效中药和中药复方及其化学成分对活性信号分子与信号转导途径影响的作用机制,对研制开发安全有效的抗肾间质纤维化中药具有重要意义。OBJECTIVE: To summarize related research of transforming growth factor beta (TGF-β)/Smad signal pathway as well as the feasibility and advantage of renal interstitial fibrosis after intervening with traditional Chinese medicine. DATA SOURCES: The Medline database was undertaken to identify the literatures of TGF-β/Smad signal pathway as well as traditional Chinese medicine against renal interstitial fibrosis published from January 1996 to June 2006 with the key words of "TGF-β/Smad signaling, traditional Chinese medicine, renal interstitial fibrosis" in English. Meanwhile, China Journal Full-text Database (CJFD) was undertaken for the same literatures published between January 2001 and June 2006 with the key words of "TGF-β/Smad signaling, traditional Chinese medicine, renal interstitial fibrosis" in Chinese. STUDY SELECTION: The data were checked primarily, and the related articles of TGF-β/Smad signal pathway as well as traditional Chinese medicine against renal interstitial fibrosis were selected. Repetitive researches were excluded. DATA EXTRACTION: A total of 54 articles of TGF-β/Smad signal pathway with traditional Chinese medicine against renal interstitial fibrosis were collected. Totally 13 trials or clinical researches were accorded with inclusive criterion. The 41 excluded articles were repetitive research. DATA SYNTHESIS: TGF-β/Smad signal pathway were the last common pathway of renal interstitial fibrosis. Intervention of TGF-β/Smad signal pathway had active significance for blocking renal interstitial fibrosis. Traditional Chinese medicine had multiple substances, and in theory it was characterized by multiple pathways and multiple targets. As to the complicated network of TGF-β/Smad signal pathway, it played an important comprehensive effect.
