摘 要:目的:探讨凋亡相关基因bcl-2在肾小管损伤及间质纤维化过程中的表达以及扶正活血化瘀中药芪红合剂在肾小管损伤及间质纤维化过程中的肾保护作用及可能的作用机制。方法:采用单侧输尿管结扎致肾间质纤维化大鼠模型,将大鼠随机分为4组:假手术组、模型组、中药芪红合剂组、依那普利组,术后10d处死各组大鼠,收集血清测定尿素氮(BUN)、血肌酐(Cr)水平,结扎侧肾组织行HE、Masson染色,Masson染色分析肾间质纤维化程度,免疫组化半定量检测各组大鼠肾组织中bcl-2、FN的蛋白表达。结果:模型组bcl-2、FN的蛋白表达及肾间质纤维化面积明显高于假手术组,BUN、Cr水平均显著升高(P<0.05);中药芪红合剂组、依那普利组较模型组bcl-2的蛋白表达显著上调,FN的蛋白表达及肾间质纤维化面积明显降低,BUN、Cr水平均显著下降(P<0.05)。结论:中药芪红合剂可能通过增强凋亡调控基因bcl-2蛋白表达的上调而减轻肾小管损伤及间质纤维化。Objective: To study the effect of Qihong decoction on bcl-2 expression in renal cortex of model rats with tubulointerstitial fibrosis. Methods: Model rats were induced by ligating unilateral ureter. 40 rats were randomized into sham operation control group, model group, Qihong decoction group, enalapril group. The sera and the kidney tissues were collected after 10 days treatment. BUN and Cr in serum were measured and the level of tubulointerstitial fibrosis was observed by HE and Masson stainings. The protein expressions of bcl-2 and fibronectin(FN)in kidney tissue were measured by immunohisto-chemistry. Results: Compared with model group, the bcl-2 expression increased obviously, the FN expression and the levels of BUN, Cr decreased significantly in Qihong decoction group and enalapril group. Conclusion:Qihong decoction could alleviate renal tubule damage and tubulointerstitial fibrosis through upregulating protein expression.
