防己茯苓汤加减对TNF-α诱导的大鼠肾小球系膜细胞增殖及PAI-1含量的影响

摘　　要：目的观察防己茯苓汤加减方是否具有干预肿瘤坏死因子(TNF-α)诱导后肾小球系膜细胞增殖的作用及对纤溶酶原激活物抑制剂(PAI-1)的影响。从而为临床运用此方治疗慢性肾脏病提供实验依据。方法采用大鼠肾小球系膜细胞培养进行体外研究,用TNF-α刺激系膜细胞增殖,再加入不同浓度防己茯苓汤加减方提取液对细胞进行干预,用MTT法测定细胞增殖,流式细胞仪分析细胞周期,用ELISA法测定细胞培养液中肾小球系膜细胞纤溶酶原激活物抑制剂(PAI-1)的含量。结果加入防己茯苓汤加减方提取液后,系膜细胞的增殖程度受到显著抑制(P<0.05),流式细胞仪DNA图示S期细胞数显著减少(P<0.05)。细胞培养液中PAI-1的表达受到抑制(P<0.05)。结论防己茯苓汤加减方对TNF-α诱导后肾小球系膜细胞的增殖具有抑制作用,并可影响肾小球系膜细胞PAI-1的表达,进而调整细胞外基质(ECM)的合成与降解。Objective To investigate the effect of Fangjifulingtang on preventing proliferation at glomerular mesangial cells and the expression of PAI-1, so as to provide experimental evidence for Fangjifulingtang to cure chronic kidney disease. Method Glomerular mesangial cells were cultured and TNF-α was used as a stimulation to make the cells proliferate. Then Fangjifulingtang was incubated together at different doses. Cell vitality were evaluated and qualified by MTT. Cell period were analysed by Flow CytoMeter （FCM）. Secretion of PAI-1 expression was measured by ELISA. Results The proliferation of Glomerular mesangial cells was inhibited （P〈0.05）. The DNA diagram of Flow CytoMeter showed that the cell quantity in stage ‘S＇ was reduced （P〈0.05） and the expression of PAI-1 was significantly inhibited （P〈0.05）. Conclusion Fangjifulingtang was able to inhibit proliferation of glomerular mesangial cells and influence the expression of PAI-1, so as to regulate the synthesis and degradation of extracellular matrix.
