摘  要:目的:观察草酸-S-西酞普兰对慢性应激抑郁大鼠模型行为学和血浆皮质醇(CORT)、促肾上腺皮质激素(ACTH)含量的影响。方法:SD雄性大鼠40只随机分为正常组、模型组、草酸-S-西酞普兰低、中、高剂量(1, 2,4mg·kg^(-1))组。给药组大鼠从造模d1起每天8:00灌胃给药,连续21d。模型组给予等体积生理氯化钠洛液。选用慢性轻度不可预见性应激加孤养造模,观察各组大鼠敞箱实验和液体消耗等行为学指标变化,采用放射免疫方法检测大鼠血浆CORT和ACTH含量。结果:与正常组比较,模型组大鼠体重增加缓慢。敞箱实验中的水平运动、垂直运动得分、理毛次数显著减少,中央格停留时间显著延长,糖水偏爱度明显下降,且其血浆CORT和ACTH含量增加。草酸-S-西酞普兰低、中、高剂量组均显著改善慢性应激抑郁大鼠模型的行为学和神经内分泌变化。结论:慢性轻度不可预见性应激可使大鼠行为及神经内分泌发生异常改变,引起抑郁状态,草酸-S-西酞普兰对此具有一定拮抗作用。Objective :To evaluate the effects of escitalopram on neuroendocfine and behavior of chronic stress depression rats. Methods: Forty male Sprague-Dawley rats were randomly allocated to five groups: normal control group,model group,three escitalopram groups. The control rats were supplemen- ted with normal saliue solution and escitalopram-treated rats were fed with eseitalopram 1 , 2 or 4 rag" kg J for 21 days. All rats except the control group are singly housed and exposed to an unpredieted sequence of mild stressor. The rats' behaviors were monitored by open-field test and sucrose preference test. The levels of plasma adrenocorticotropic hormone (ACTH) and cortisol (CORT) were measured by radioimmunoassay. Results:Chronic unpredicted mild stress affected rats neuroendoerine and behaviors, causing depression. Compared with normal control rats,the model rats showed significantly decreased body weight and experienced less squares crossed, rears and grooming activities ill the open-field test. Sucrose consumption in the sucrose preference test decreased significantly. The conceutration of plasma ACTH and CORT significautly increased in stress-model rats compared to the control rats. The escitalopram-treated rats signifieantly improved their behavioral disorders and the levels of plasma ACTH and CORT. Conclusion:Escitaloppram antagonized HPA hyperactivity of rats, suggesting anti-depressant activity.
