维拉帕米对柯萨奇B_3病毒感染培养心肌细胞的作用

摘　　要：目的：研究维拉帕米对柯萨奇Ｂ３病毒（ＣＶＢ３）感染新生大鼠培养心肌细胞离体模型的影响。方法：制备ＣＶＢ３感染培养的新生ＳＤ（Ｓｐｒａｇｕｅ－Ｄａｗｌｅｙ）大鼠搏动心肌细胞。观察细胞病变，在维拉帕米实验组及生理盐水对照组检测培养液中心肌型肌酸激酶同功酶，培养液及细胞悬液中ＣＶＢ３－核糖核酸（ＣＶＢ３－ＲＮＡ），另设实验研究培养心肌细胞钙离子流入量。结果：ＣＶＢ３感染经维拉帕米处理组（５×１０－７及５×１０－８ｍｏｌ／Ｌ，ｎ＝８）细胞病变较生理盐水处理组（ｎ＝８）、地塞米松处理组（１及２×１０－４ｍｏｌ／Ｌ，ｎ＝８）及人体重组干扰素α１组（１０２及１０３Ｕ／Ｌ，ｎ＝８）明显严重（Ｐ均＜０．０１）；生理盐水处理组心肌型肌酸激酶同功酶显著低于维拉帕米处理组（Ｐ＜０．０１）。生理盐水处理组细胞悬液中总核糖核酸与ＣＥＶ２－互补脱氧核糖核酸探针杂交带小于维拉帕米处理组；生理盐水处理组培养液中总核糖核酸与ＣＥＶ２－互补核糖核酸杂交带小于维拉帕米处理组，杂交指数小于维拉帕米处理组（Ｐ＜０．０５）。ＣＶＢ３感染使培养心肌细胞的快速及慢速时相钙内流明显增加（（Ｐ＜０．０５），维拉帕米可减低慢速时相钙内流（Ｐ＜０．０１），但对快速?Objective:To study the effect of verapamil (VR) on Coxsackievirus B 3(CVB 3)infected newborn rat myocardial cells in culture. Methods:CVB 3 infected cultured myocardial cells of newborn SD rat were prepared,and the effect of VR on these cells was studied by examination of cytopathic changes (CPE),MB creatine kinase (CK MB),amount of CVB 3 RNA,and cellular calcium influx(Ca influx). Results:The CPE of cultured cardiac cells was significantly aggravated in VR(5×10 -7 and 5×10 -8 mol/L, n =8) treated groups compared with control (normal saline,NS, n =8),dexamethasone(Dex,l and 2×10 -4 mol/L),and recombinant human interferon α 1(rHu IFN α 1,10 2 and 10 3U/L, n =8)groups (all p <0.01).Compared with the NS group, CK MB activity( n =8) in the culture medium,and CVB 3 RNA in both the culture medium and cells ( n =8) by semi quantitative dot blot hybridization were significantly higher ( p <0.01 and 0.05,respectively) in the VR treated group.Both the rapid and slow phases Ca influx of cultured cardiac cells were markedly increased ( p <0.05) by CVB 3.VR decreased Ca influx at the slow phase ( p <0.01),but had no effect at the rapid phase. Conclusion:VR aggravates myocardial pathologic changes caused by CVB 3, and accelerates the replication of CVB 3 in the myocardium. VR should be used in patients of CVB 3 myocarditis with caution.
