摘 要:目的:研究辣椒素受体(TRPV1)在内皮素(ET1)引起内脏痛中的作用。方法:选用TRPV1基因敲除小鼠(KO小鼠)及其野生型C57BL/6小鼠(WT小鼠),均接受腹腔内注射0.6%醋酸(10ml/kg,n=6)和硫酸镁(60mg/kg,n=6),以及按50、25、12.5、4μg/kg注射ET1(每组每一剂量n=10),注射后观察小鼠的行为,记录扭体反应的情况。结果:WT小鼠在硫酸镁或醋酸腹腔内注射后均引起扭体,而KO小鼠没有出现扭体。两组注射ET1后均出现扭体,呈剂量依赖性,但WT组在各剂量时扭体反应发生率均明显高于KO组,扭体次数在剂量为50、25和12.5μg/kg时明显高于KO组。结论:腹腔注射ET1能够引起内脏痛,其机制之一在于激活了TRPV1。Objective:To investigate the role of transient receptor potential vanilloid subtype 1 (TRPV1) in the nociceptive response induced by intraperitoneal endothelin-1 ( ET-1 ). Methods : Magnesium sulfate (60 mg/kg) and 0.6% acetic acid (10 ml/kg) were intraperitoneally injected in TRPV1 gene knockout mice (KO group, n =6) and wildtype mice (WT group, n =6). ET-1 at the doses of 50, 25, 12.5 and 4 μg/kg were injected in both groups(n = 10 at each dose). The behavior and writhing response of the mice were observed for 30 minutes. Results: Magnesium sulfate or acetic acid induced writhing in WT mice but not in KO mice. ET-1 induced dose-dependent writhing in both KO and WT mice, but the response rate of writhing was lower in KO group than in WT group at all doses. And writhing times of KO group were less than those in WT group at doses of 50, 25 and 12.5 μg/kg. Conclusion : Intraperitoneal injection of ET-1 caused visceral pain. TRPVI may be involved in the mechanisms of nociception elicited by ET-1.
