单克隆天然抗角蛋白抗体3B4抑制白念珠菌芽管生成的研究

摘　　要：目的观察单克隆天然抗角蛋白抗体384与白念珠菌结合及对芽管生成的影响。方法采用ELISA和流式细胞仪检测单克隆天然抗角蛋白抗体384与白念珠菌结合,并将抗体与真菌共孵育,检测白念珠菌芽管生成的变化。结果ELISA和流式细胞仪检测均发现3B4可以与白念珠菌结合；相对于同型对照组,384作用组真菌芽管生成率显著下降,两组差异有统计学意义。结论单克隆天然抗角蛋白抗体3B4可以与白念珠菌结合并抑制其芽管生成,提示天然IgM在机体抗真菌天然免疫中可能具有一定的作用。Objective To investigate the effect of the binding of a natural monoclonal antikeratin antibody, 3B4, to Candida albicans on germination. Methods The binding of 3B4 to C. albicans was analyzed by ELISA and flow cytometry （ FCM ）. The effect of 3B4 on germination was analyzed by germ tube forming assay. Results Both ELISA and FCM showed binding of 3B4 to C. albicans. Germ tube forming assay found that the germination rate significantly decreased in 3B4-added group compared to the control group with the same type of antibody added. Conclusion The natural monoclonal antikeratin antibody 3B4 can bind to C. albicans and inhibit its germination, which indicates that natural antikeratin antibody has some role in the natural immunity against yeast infection.
