摘 要:目的探讨高强度超声聚焦(HIFU)抗大鼠早孕的有效剂量及可能机理。方法应用HIFU以14、12、10、8及6W/cm25种不同声强辐照孕8d大鼠左侧子宫角胚胎着床点,以右侧为对照。孕15d观察计算胚胎死亡率。同时用TUNEL染色检测子宫蜕膜细胞凋亡率。结果5种声强致大鼠胚胎死亡率与对照侧有显著差异(P<0.05)。声强≥10W/cm2与<10W/cm2胚胎死亡率有显著差异(P<0.01)。辐照侧蜕膜细胞凋亡指数与对照侧差异明显(P<0.05)。结论HIFU对大鼠胚胎完全性致死的剂量阈值为10W/cm2×90s。蜕膜细胞凋亡参与了HIFU抗大鼠早孕的过程。Objective To study the anti-gestational effects of high intensity focused ultrasound. Methods Female rats of 8 day gestational age were anesthetized and their uterine horns were exteriorized through abdominal incision. The embryo near the left oviduct was exposed to ultrasound irradiation and the right corresponding one served as the control. The rats were exteriorized for examination on the 15th gestational day. The decidua of both normal pregnant and abortion rats were scratched for TUNEL (terminal deoxynucleotity transferase mediated dUTP nick end labeling) detection. Results The embryo dead rate of 14 W/cm^2, 12 W/cm^2, 10 W/cm^2, 8 W/cm^2 and 6 W/cm^2 of HIFU were100 %, 100 %, 100 %, 72.86 % and 36.21% respectively. The apoptosis ratio of the experimental group was significantly higher than that of the control group, P 〈 0.05. Conclusions The embryos exposed at 14 W/cm^2, 12 W/cm^2 and 10 W/cm^2 of HIFU on 8th gestational day were 100% dead. Therefore, HIFU may induce the apoptosis of decidual cells.
