摘  要:目的通过不同的原料和生化提取方法,制备了小牛血清去蛋白注射液(DCSI)和小牛血去蛋白注射液(DCBI),比较两者在主要成分、多肽含量、生物活性等方面的差异。方法将DCSI及DCBI制备好后,对两者进行试验对比研究,即采用高效液相色谱法进行鉴定分析,用Lowry法测定多肽含量,用瓦氏微量呼吸检压仪测定呼吸活力并计算刺激指数,测定两者对小鼠耐缺氧试验的不同影响时间。结果DCSI与DCBI相比,前者色谱的双峰面积大于总的峰面积75%,显著区别于后者的60%,并且前者在多肽含量,呼吸活性及小鼠耐缺氧时间的指标上都明显高于后者(P<0.05)。结论DCSI与DCBI之间的主要成分、主要成分的含量和生物活性有显著的差别,并且前者优于后者,从药物分析角度说明两者不是同一种产品。Objective To observe the differences in the polypeptide and active ingredient contents and biological activity between deproteinized calf serum injection (DCSI) and deproteinized calf blood injection (DCBI). Methods DCSI and DCBI, prepared with different materials and biochemical extraction methods, were identified with high performance liquid chromatography (HPLC) and compared in the content of active ingredient with the Lowry' s way, and in the cellular respiration and stimulation index with the method ofmicrorespiromete ation decreasing. In addition, the effect of the 2 injections on the rat tolerance to anoxia was observed. Results Double-peak area accounted for more than 75% of total peak area in HPLC of DCSI, which was significantly higher than that (about 60%) in HPLC of DCBI; DCSI not only prolonged the duration of tolerance to anoxia as compared with DCBI, but also had higher biological activity and polypeptide content (P〈0.05). Conclusion There are obvious differences in the contents of active components and biological activity between DCSI and DCBI, and DCSI is better than DCBI.
