摘  要:目的:观察大鼠右侧腓总神经损伤局部植入酸性成纤维细胞生长因子纤维蛋白凝胶载体后的形态学及酶组织化学的变化。方法:实验于2005-07/10在南方医科大学珠江医院完成。①选取清洁级SD大鼠30只,随机分为3组:酸性成纤维细胞生长因子+纤维蛋白凝胶载体组、单纯纤维蛋白凝胶载体组、空白对照组,10只/组。②各组大鼠全麻状态下均切断右侧腓总神经,距神经入肌点约1cm处。酸性成纤维细胞生长因子+纤维蛋白凝胶载体组缝合神经外膜,失神经支配的胫前肌神经区周围的肌间隙植入圆柱形复合物(含酸性成纤维细胞生长因子25μg,纤维蛋白凝胶载体7.5mg);单纯纤维蛋白凝胶载体组缝合神经外膜,失神经支配的胫前肌神经区周围的肌间隙单纯植入纤维蛋白凝胶载体7.5mg;空白对照组缝合神经外膜但不给药。③各组大鼠全麻下对称显露双侧胫前肌,行右侧(处理侧)与左侧(正常侧)对比,观察右侧胫前肌外观变化情况,并记录各组右侧胫前肌湿质量。6周后取材进行酶组织化学指标检测。结果:实验选取SD大鼠30只,全部进入结果分析。①各组胫前肌大体形态学观察结果:酸性成纤维细胞生长因子+纤维蛋白凝胶载体组两侧胫前肌对称且无明显肌萎缩,肌张力正常;单纯纤维蛋白凝胶载体组及空白对照组右侧胫前肌均明显萎缩,肌肉硬度增加,肌张力下降。②各组肌肉湿质量检测结果的比较:与单纯纤维蛋白凝胶载体组、空白对照组比较,酸性成纤维细胞生长因子+纤维蛋白凝胶载体组肌肉湿质量明显增大(0.82,0.81,1.26,P<0.001)。③各组乙酰胆碱酯酶活性检测结果:酸性成纤维细胞生长因子+纤维蛋白凝胶载体组乙酰胆碱酯酶活性强,接近于正常侧,染色均匀,运动终板结构规则;单纯纤维蛋白凝胶载体组及空白对照组乙酰胆碱酯酶活性弱,染色不均,运动终板结构松散、周边不规则。结�AIM: To observe the morphological and enzymohistochemical change of the common peroneal nerve injury region after implantation of acidic fibroblast growth factor and fibrin gel carrier. METHODS: This experiment was carried out in Zhujiang Hospital, Nanfang Medical University from July to October 2005. ① Totally 30 clean grade SD rats were randomly divided into 3 groups: acidic fibroblast growth factor+fibrin gel cartier group, simple fibrin gel cartier group and blank control group with 10 rats in each one. ② Under the general anesthesia, right common peroneal nerves of rats in each group were cut off 1 cm away from the nerve muscle entry point. In the acidic fibroblast growth factor +fibrin gel carrier group, perilemma was sutured and column aspect compound was implanted into spatium intermusculare around denervated tibial muscle nerve region (containing acidic fibroblast growth factor 25 μg and fibrin gel carrier 7.5 mg ); In the simple fibrin gel carrier group, perilemma was sutured and fibrin gel carrier 7.5 mg was implanted into the spatium intermusculare around denervated tibial muscle nerve region; perilemma was sutured without medication in blank control group. ③ Under general anesthenia, bilateral tibial muscle was expose . Comparison was performed between right side (treated side) and left side (intact side), appearance change of right tibial muscle was observed and wet weight of right tibial muscle was recorded in each group. Enzymohistochemical index was detected following sample collecting 6 weeks later. RESULTS: Totally 30 rats were enrolled in the experimental group, and all of them entered the stage of result analysis. ① Gross morphological observation of tibial muscle : Bilateral tibial muscle was symmetry, there was no obvious muscular atrophy and muscular tension was normal in the acidic fibroblast growth factor +fibrin gel carrier group; Right tibial muscle was obviously shrinked , muscular rigidity was increased and muscular tension decreased in the si
