四硫钼酸盐对大鼠颈动脉内膜剥脱术后血管细胞黏附分子表达的抑制

摘  要:目的:观察铜鳌合剂四硫钼酸胺对大鼠颈动脉内膜剥脱术后内膜增生和血管壁血管细胞黏附分子表达的影响。方法:实验于2005-03/2006-01在解放军第四军医大学心血管内科实验室完成。①选用清洁级SD雄性大鼠30只,随机数字表法分为3组:血浆铜蓝蛋白(50±5)%水平组、血浆铜蓝蛋白(20±5)%水平组、未服药组,10只/组。以未服药组右侧颈总动脉作为空白对照。②以血浆铜蓝蛋白水平作为铜水平的代表,基础水平的平均值是(3.8±1.1)单位。血浆铜蓝蛋白(50±5)%水平组术前1周给予2g/L的四硫钼酸胺10mg/kg强饲,使血浆铜蓝蛋白水平在术前降低到基础水平的(50±5)%。血浆铜蓝蛋白(20±5)%水平组术前2周给予2g/L的四硫钼酸胺10mg/kg强饲,使血浆铜蓝蛋白水平在术前降低到基础水平的(20±5)%。两组均给药至术后14d。未服药组仅给予等量蒸馏水强饲。③各组均建立颈总动脉内膜剥脱模型,于术后14d深麻取材,左侧颈总动脉液氮冻存或用40g/L甲醛固定。④光镜下观察血管损伤后内膜增殖的形态学变化。WesternBlot法检测血管细胞黏附分子蛋白的表达水平。结果:实验选用清洁级SD雄性大鼠30只,全部进入结果分析。①铜缺乏对血管损伤后内膜增殖的形态学影响:血管球囊损伤术后14d,未服药组血管内膜呈弥漫性增厚,而血浆铜蓝蛋白(50±5)%水平组、血浆铜蓝蛋白(20±5)%水平组血管内膜增生程度均显著缩小。②铜缺乏对血管细胞黏附分子蛋白表达的影响:术后14d,未服药组损伤颈动脉组织的血管细胞黏附分子蛋白表达显著增高,血浆铜蓝蛋白(50±5)%水平组、血浆铜蓝蛋白(20±5)%水平组损伤颈动脉组织的血管细胞黏附分子表达均被显著抑制(P<0.01)。结论:四硫钼酸盐诱导的适度铜缺乏能够显著抑制血管细胞黏附分子的表达,这可能是四硫钼酸盐抑制血管球囊损伤术后再狭窄的原因之一。AIM: To observe the effect of tetrathiomolybdate (TM) on endomembrane hyperplasy and expression of vascular cell adhesion molecule-1 (VCAM-1) in carotid arteries of rats after balloon-injury. METHODS: The experiment was conducted in the laboratory of Department of Cardiovascular Medicine, Fourth Military Medical University of Chinese PLA between March 2005 and January 2006. ①Thirty clean grade male Sprague-Dawley rats were selected and randomly divided into 3 groups: ceruloplasmin (CPL) (50+5)% group, CPL (20±5)% group and normal group with 10 rats in each group. The right common carotid artery of the normal group served as blank control group. ②The CPL level was regarded as the copper level, the mean value of basic level was (3.8±1.1) unit. The CPL (50±5)% group was fed with the diet containing 2 g/L TM 10 mg/kg 1 week before operation to decrease the CPL level to (50±5)% of the basic level, while the CPL (20±5)% group was fed with the diet containing 2 g/L TM 10 mg/kg 2 weeks before operation to decrease the CPL level to (20±5)% of the basic level. Both of the groups were given administration until the 14a day after operation. The normal group was only given the same dosage of distilled water diet. ③All rats were subjected to balloon injury and killed 14 days later. The left common carotid artery was frozen with liquid nitrogen or fixed by 40 g/L formaldehyde." ④The morphological changes of endomembrane hyperplasy after injured was observed with the light microscope. Western blotting was used to detect the expression of VCAM-1. RESULTS: All the 30 SD rats were involved in the result analysis. ① Effect of copper deficiency on morphology of endomembrane hyperplasy after vessel injury: 14 days after balloon-injury, the endangium of the normal group presented diffused thickening, . but the hyperplasy degree of the CPL (50±5)% group and the CPL (20±5)% group were obviously lighter. ②Effect of copper deficiency on expression of VCAM
