摘  要:目的对我国登革2型病毒1993年广东省佛山市流行株GD06/93结构蛋白基因进行序列测定及分析,追踪其地域来源、基因组结构与致病性的关系,为研究开发登革病毒新型疫苗奠定基础。方法根据登革2型国际标准株NGC序列,设计2对重叠引物,RT-PCR扩增,分别克隆到pMD18-T载体,转化受体菌DH5α,挑取阳性克隆进行PCR、酶切鉴定及序列测定。结果GD06/93株结构基因序列长度为2325bp,编码775个氨基酸。与其它登革2型病毒株TSV01、Taiwan-1008DHF、NGC、44、ThD2_0038_74、ThNH81/93、gd08/98、04、Cuba165/97及S1进行比较,其核酸序列同源性分别为97%、96%、95%、95%、95%、94%、94%、92%、92%、91%。结论GD06/93病毒株的结构基因序列与已发表的其它登革2型病毒株同源性很高。在比较的6株登革2型病毒株中,GD06/93株与同期在澳大利亚流行的TSV01株同源性最高。Objective To sequence and analyze the structural gene of dengue virus type 2 strain GD06/93 isolated from Foshan, Guangdong province, in order to understand the molecular epidemiology of dengue virus and the development of dengue virus vaccine. Method Two pairs of primers were designed according to the sequence of dengue virus type 2 strain NGC. The gene coding for the structural protein gene was amplified by reverse transcriptase-polymerase chain reaction (RT-PCR). The gene was then cloned into pMD18-T vector and sequenced. Result The isolated structural gene had a total of 2325 base pairs and encoding for 775 amino acids. The sequence homology of this structural gene with genes from other strains was: strain TSV01(Australia 1993), 97%; strain Taiwan-1008DHF, 96%; strain 44 (China), ThD2_0038 74(Thailand) and NGC (New Guinea-C) 95%; strain gd08/98 (China) and ThNH81/93(Thailand 1993), 94%; strain 04 (China) and Cuba165/97 (Cuba, 1997), 92%; and strain S1,91%. Conclusion The sequence of the structural gene is identical to the published gene in dengue virus type 2. The gene shows a high degree of sequence homology with TSV01, and a relative low sequence homology with S1.
