摘  要:目的:探讨人类免疫缺陷病毒(human immunodeficiency virus,HIV)gp120对体外培养的背根神经节神经元胞体和突起生长的影响。方法:分散培养的胎鼠背根神经节神经元用不同浓度的HIVgp120(125、250、500 pmol/L和1 nmol/L)处理,对分散培养的背根神经节神经元用MAP2免疫荧光标记后,用共聚焦激光扫描显微镜观察神经元胞体和突起的改变。结果:应用gp120后7 d,神经元突起的数目减少,长度变短,而神经元的胞体则没有明显变化。结论:HIV gp120可直接影响分散培养的背根神经节神经元突起的生长。Objective: To determine the effects of human immunodeficiency virus (HIV) gp120 on dissociated cttltured dorsal root ganglion(DRG) neuronal growth. Methods: Dissociated cultured rat embryo's DRG cells were treated with HIV gp120 at different concentration (125 pmol/L, 250 pmol/L, 500 pmol/L, 1 nmol/L, respectively) and then were processed for MAP2 immunofluorescent labeling, and furthermore the changes of neuronal cell body and neurites were observed with confocal laser scanning microscope. Results: The number and length of neurites were reduced after 7-day treatment with gp120 compared with control group, whereas the diameter of neuronal cell body was not obviously altered compared with control group. Conclusion: HIV gp120 may have direct influences on the outgrowth of neurites in dissociated DRG cell cultures.
