白花前胡提取液含药血清对心肌细胞生长的影响

摘　　要：目的:探讨在缺乏过度生长刺激条件下白花前胡提取液含药血清对体外培养正常心肌细胞生长的影响。方法:采用免疫沉淀技术、免疫细胞化学染色及流式细胞仪观察使用白花前胡含药血清前后心肌细胞生长情况及搏动频率、原癌基因表达、细胞周期变化。结果:白花前胡提取液含药血清高低剂量组孵育24h,心肌细胞贴壁伸展状态均良好,未见明显细胞脱落、悬浮现象,心肌细胞搏动频率与空白组无显著差异(P>0.05),使用前胡提取液高低剂量含药血清2h后对心肌细胞c-Jun蛋白表达及其在细胞内的分布无显著影响(P>0.05),白花前胡提取液含药血清高低剂量组心肌细胞G0/G1期、S期、G2/M期分布比率均与空白组无显著性差异(P>0.05)。结论:白花前胡提取液含药血清对正常心肌细胞的活力无显著影响,不干扰正常心肌细胞的细胞信号传导过程,对心肌细胞周期无明显影响。Objective：To study the effect of Pd-E contained serum on cultured cardiomyocytes without excessive growth stimulation. Method：MTT,The methods of immunocytochemical staining and flow cytometry were used to observe the growth,pulsating frequency,the expression of proto-oncogene and the cell cycle of cardiomyocytes. Results ：The growth,pulsating frequency ,the expression of c-jun and the cell cycle of cardiomyocytes in the Pd-E stimulating group have no obvious difference with the control group （P 〉 0.05）. Conclusion： Pd-E contained serum has no significant effect on the normal cardiomyocytes and the signal transduction and the cell cycle of them.
