摘 要:目的观察醒脑静对内毒素所致大鼠全身炎症反应综合征(SIRS)中的作用。方法静脉注射脂多糖(LPS)建立大鼠SIRS模型。雄性Wistar大鼠85只,随机分为假手术对照组(C组)、LPS模型组(LPS组)和醒脑静治疗组(XNJ组);XNJ组依据给药剂量不同(5,10,15 mL.kg-1)又分为XNJ-5,XNJ-10,XNJ-15共3个亚组。LPS组和XMJ组按观察时间分为1,2,4,6 h 4个亚组。运用酶联免疫吸附分析(ELISA)法检测单个核细胞中NF-κB活性、血清中TNF-α及IL-6水平,并观察肺脏和肝脏的病理学变化。结果LPS组单个核细胞中NF-κB活性及TNF-α浓度明显升高,2 h最明显,显著高于对照组;血清IL-6浓度随着时间的推移不断升高,显著高于对照组,病理结果显示,LPS组肺泡出血、水肿、大量炎症细胞浸润,肝脏毛细血管扩张、充血、水肿和炎症细胞浸润。XNJ组与LPS组比较,NF-κB活性、TNF-α及IL-6水平显著降低,肺脏和肝脏的病理损伤减轻。结论醒脑静可通过抑制NF-кB活性、TNF-α及IL-6水平,改善内毒素所致大鼠全身炎症反应综合征。OBJECTIVE To investigate the role of Xingnaojing injection in rats with systemic inflammatory response syndrome (SIRS). METHODS The model of SIRS rats were induced by infusion LPS through vein. Eighty-five male Wistar rats were randomly divided into 3 groups, the control(C) group, the LPS group and the Xingnaojing(XNJ) group. According to the difference of dosage(5,10,15 mL·kg^-1), the Xingnaojing group was divided into 3 subgroups, the XNJ-5 subgroup, the XNJ-10 subgroup and the XNJ-15 subgroup. The LPS group and the Xingnaojing group were divided into 4 subgroups which was 1, 2, 4 and 6 h subgroup according to different time. At 1,2,4 and 6 h after LPS injection, the activation of NF-κB in blood mononuclear cells, the content of TNF-α and IL-6 in plasma were detected by enzyme-linked immunoadsordent assay(ELISA).The pathological changes of lung and liver tissue were also examined. RESULTS After LPS injection, the activation of NF-κB in mononuclear cells and the level of TNF-α increased clearly and reached its top at 2 h in LPS group, which was higher than that of control group after LPS infusion. The content of IL-6 was increased gradually with time in LPS group, which was higher than that of control group after LPS injection. Pathological examination showed that LPS caused pulmonary alveolar hemorrhage, edema and inflammatory cell infiltration in lung tissue and congestion, edema, capillary dilation and inflammatory cell infdtration in liver tissue in LPS group. Compared with LPS group, the activation of NF-κB, the levels of TNF-α and IL-6 decreased,and the damage of lung and liver tissue was relieved in Xing-naojing group. CONCLUSION Xingnaojing injection improves endotoxin induced SIRS by inhibiting the activity of NF-κB, the levels of TNF-α and IL-6.
