摘  要:目的:观察不同大小的+Gz暴露对大鼠下丘脑部位HSP 70 mRNA表达水平的影响,探讨+Gz暴露致脑损伤的机理。方法:100只SD大鼠随机分为对照组,+2 Gz暴露组,+6 Gz暴露组,+10 Gz暴露组,各组在G暴露后的5 h、10h1、d、2 d、4 d处死,采用原位分子杂交方法观察大鼠脑下丘脑部位HSP 70 mRNA表达情况,并对实验结果进行统计分析。结果:3个+Gz暴露组,HSP 70 mRNA表达水平表现出相同的变化趋势:在+Gz暴露后5 h,HSP 70 mRNA表达水平开始增加,10 h达峰值,4 d时基本恢复正常,而对照组没有这一变化趋势,不同G值之间的比较表明,+6 Gz组HSP 70 mRNA表达水平最高,+10 Gz组最低。结论:+Gz暴露对大鼠下丘脑部位HSP 70 mRNA表达水平有着显著的影响。Objective:To observe the changes of expression of HSP 70 mRNA in hypothalamus of rats after they were exposed to +Gz of different magnitudes and investigate the mechanism of brain injury induced by +Gz exposure. Methods. 100 rats were divided into 4 groups: control group, +2 Gz group, +6 Gz group and +10 Gz group. The experimental animals underwent +2 Gz, +6 Gz and +10 Gz exposure for 3 min, respectively. The animals were killed at 5 h, 10 h, 1 d, 2 d and 4 d after the exposure, their hypothalamus specimens were got and the levels of expression of HSP 70 mRNA were determined by means of situ hybridization technology. Results. The levels of expression of HSP 70 mRNA in the experimental groups were higher than that of the control group. It began to rise at 5 h after the exposure, reached to peak at 10 h, and returned to the normal at 4 d after the exposure. Among 3 experimental groups, the expression level was highest in +6 Gz group and lowest in +10 Gz group. Conclusion. +Gz exposure can induce obvious changes of the expression of HSP 70 mRNA in hypothalamus of rats.
