摘 要:目的选择适当剂量γ射线辐照机采单个供者血小板(SDP),不损伤SDP且预防输血相关移植物抗宿主病(TA-GVHD)发生。方法选择13份SDP样本并均分为3组,2组分别选择30Gy和40Gy的γ射线辐照,未辐照且同温度条件下样本为对照组,检测乳酸脱氢酶(LDH)值等并比较。结果3组间LDH检测均无显著性差异,血小板形态、聚集程度等均无显著性变化,pH检测符合SDP保存条件。结论LDH等相关检测表明,选择30-40Gy辐照剂量对SDP无显著性损伤,灭活淋巴细胞效果显著且时间较短。Objective To select optimal dose of γ-ray to radiate single donator platelet (SDP) for prevention TA-GVHD and protect platelet from being injured.Methods 13 samples of SDP were selected and radiated by 2 dose(30Gy, 40Gy) of γ-ray, then the levels of lactate dehydrogenase(LDH) were measured and compared with those of control group. Results No significant difference of the LDH levels was shown between radiated group and control group,and no significant difference was found between 2 radiated groups.No significant variation was shown of the shape of platelet and their congregation extent. The value of pH was proper for preserving SDP.Conclusion Radiation dose from 30-40Gy is suitable for SDP without being injured and effective for deactivation of lymphocyte.
