摘  要:为探讨小黑麦抗旱的生理特性及分子机理,选用小黑麦H01-4,在30%PEG-6000室内模拟干旱条件下,分别对其抗旱指标叶片相对含水量和脯氨酸进行测定,并利用mRNA差异显示方法分析小黑麦幼苗叶片基因表达差异,结果表明:随着胁迫时间的延长,叶片相对含水量呈下降趋势,复水后上升,再胁迫下降;而脯氨酸的含量变化则与叶片相对含水量呈相反趋势变化;小黑麦幼苗叶片基因表达存在数量水平和质量水平的差异。量的表达差异表现为随着胁迫的加强表现为增强或减弱;质的表达差异表现为有新带的产生或消失。此外,对影响mRNA差异显示技术的一些因素也作了讨论。In order to study the physiological property and the molecular mechanism of droughtresistance in triticale , under 30%PEG - 6000 simulation drought condition, triticale H01 - 4 was selected, indicator leaf relative water content(RWC) and proline, were determined respectively and the differences in gene expression in seedling of triticale by using rnRNA differential display were analyzed. The results indicated that RWC descend along with extension of time stress, ascend after rehydration , and descend after more stress; but change of proline content show an opposite tendency against RWC; The patterns of gene expression differ significantly In seedling of triticale . Both quantitative and qualitative differences were found, The quantitative differences show gene overexpression, underexpression along with stress reinforce; The qualitative differences show appearance or disappearance of new belt. Besides, several factors about DD - PCR were also discussed.
