摘 要:目的测定不同采收期川黄柏雌、雄株中的小檗碱和黄柏碱含量,探索小檗碱和黄柏碱在不同采收期的含量变化规律。方法采用RP—HPLC法,选用Luna C18(2)色谱柱(6.0mm×150.0mm,5μm);测定小檗碱的流动相为甲醇-磷酸缓冲液-十二烷基硫酸钠(60:40:0.1),检测波长350nm,流速1ml·min^-1,柱温30℃。测定黄柏碱的流动相为乙腈-0.4mol·L^-1氯化氨(12:88),检测波长286nm,流速1ml·min^-1,柱温30℃。结果雌株中小檗碱、黄柏碱的含量在6-8月份含量降低,而雄株中两者无明显变化。结论川黄柏雌株在6~8月小檗碱、黄柏碱含量骤降,与川黄柏的花果期密切相关。OBJECTIVE To determine the contents of berberine and phellodendrine in Phellodendron chinese from different collection periods and to discover the inherent rule about the content change owing to different collection periods. METHODS Luna C18 (2) ( 6.0mm×150.0mm,5μm) clumn was used for the detection,with flow rate of 1 ml·min^-1 and column temperature of 30℃. The mobile phase berberine and phellodendrine were methanol - phosphate buffer - sodium dodecylphate (60:40:0. 1 ) and acetonitrile - 0. 4 mol· L^-1 ammonium chloride(12:88) with detection wavelength of 350 m and 286 nm,respectively. RESULTS The contents of berberine and phellodendrine in female plants decreased in June,July, and Augest,but not in the male plants. CONCLUSION The contents of berberine and phellodendrine dramatically decreased in June,July and Augest,which was highly related to the time of blossom and early time of fructification.
