中国荷斯坦奶牛CSN1S2基因第二外显子SSCP多态性与产奶性状的关系

摘　　要：利用PCR-SSCP分子标记技术,研究了中国荷斯坦奶牛sα2-酪蛋白基因(CSN 1S 2基因)第二外显子的多态性,并分析了其与产奶性状之间的相关性。结果表明,中国荷斯坦奶牛CSN 1S 2基因第二外显子存在A和B2个等位基因,有AA、BB和AB 3种基因型,A和B的等位基因频率分别为36.54%和63.46%,AA、BB、AB基因型的频率分别为19.2%,46.1%和34.6%,群体中多态信息含量为0.356 3,达中度多态性,遗传变异较大;CSN 1S 2基因第二外显子对产奶量和乳脂含量没有明显影响,但AA基因型个体乳蛋白含量显著高于BB和AB基因型个体(P<0.05),表明A基因对中国荷斯坦奶牛乳蛋白含量有明显影响。The polymorphism of CSN1S2 gene exon2 of the China Holstein were studied through PCRSSCP （polymerase chain reaction-single strand conformation polymorphism）in this reasch. The result showed.there were two alleles A and B,three genotypes AA,BB and AB of the exon2 of CSN1S2 gene. The gene frequencies of A and B were 36. 54% and 63.46% separately. The genotype frequencies were 19. 2% ,46.1% and 34.6% respectively. The PIC （polymorphism information content） was 0. 356 3,indicating the exon2 of CSN1S2 gene of the China Holstein had high polymorphism and large variation. The exon2 of CSN1S2 gene had no significant influence on milk yield and creaminess. The cow with genotype AA had more lactoprotein than the cow with genotype BB and AB（P〈0.05）. So gene A has significant influence on lactoprotein of the China Holstein.
