摘  要:将大鼠肠粘膜微血管内皮细胞分为空白对照组、BSA对照组、LPS1组、LPS2组、LT 1组和LT 2组6个组,分别用维持培养液,含50μg/mL BSA、1或10μg/mL LPS、10或50μg/mL LT的培养液静置培养12 h,研究不同浓度LPS和LT对体外大鼠肠粘膜微血管内皮细胞形态及NO分泌量的影响,并确定LPS和LT致大鼠肠粘膜微血管内皮细胞损伤的模型剂量。结果表明,LPS可使细胞长宽比增大,间隙变宽,而LT可使细胞肿大,排列紊乱;各浓度的LPS和LT均能引起肠粘膜微血管内皮细胞的NO分泌量升高,但1μg/mL LPS与50μg/mL LT的作用更明显;LPS和LT致大鼠肠粘膜微血管内皮细胞损伤的模型剂量分别是1和50μg/mL。The intestinal mucosa microvascular endothelial cells were divided into blank control group, BSA control group, LPS1 group, LPS2 group, LT1 group and LT2 group. By adding feeding medium 50 μg/mL BSA, 1 μg/mL LPS, 10 μg/mL LPS,10 μg/mL LT and 50 μg/mL LT culture fluid respectively into the 6 groups to cultivate statically for 12 hours,the respective effects of different concentrations of LPS and LT on the intestinal mucous membrane microvascular endothelial cells' shape and excreting NO was studied,then the model dosage of LPS and LT damaging intestinal mucosa microvascular endothelial cells in rats was decided. LPS makes the ratio between the length and the width of a cell increase and the gaps between cells wider;LT makes the cells swell and the alignment in disorder. Both LPS and LT of every concentration can lead to the rising of the secretion of NO by intestinal mucosa microvascular endothelial cells, but the function of 1 μg/mL LPS and 50 μg/mL LT is more obvious. The model dosage of LPS and LT damaging intestinal mucosa microvascular endothelial cells in rats is respectively 1μg/mL and 50μg/mL.
