摘  要:目的观察榄香烯(Ele)、姜黄素(Cur)对体外培养的牛晶状体上皮细胞(LECs)凋亡的超微结构的影响。方法分别用160μg/ml Ele,20μg/ml Cur与LECs共同孵育24、48、72h,采用透射电子显微镜观察LECs超微结构的变化。结果Ele和Cur作用不同时间的LECs均出现核染色质凝集、固缩、边集等典型的凋亡形态学改变,呈时-效依赖关系。超微结构研究显示,Cur诱导凋亡的同时,也诱导发生细胞胀亡。结论Ele和Cur具有明显诱导LECs凋亡或胀亡的作用。Objective Elemene(Ele) and curcumin(Cur) can induce the apoptosis of various cancer cells. However,their mechanism of action on lens epithelial cells (LECs) are unknown. In the present study, we investigated the uhrastructural features of apoptosis of cultured LECs induced by Ele and Cur. Methods LECs were incubated with DMEM medium + 160 μg/ml Ele ( Ele group) or DMEM medium + 20 μg/ml Cur( Cur group) or DMEM medium( blank medium) for 24,48,72 hours separately. 1 × 10^6/ml of cultured cells were collected. The uhrastructure of incubated LECs was observed by transmission electron microscope. Results The typical apoptosis of incubated LECs with Ele and Cur was observed under transmission electron microscope. Different stages of apoptotic cells were seen in Ele group,showing heterochromatin condensation and aggregated at the periphery of cell nuclear, condensed chromatin with many vacuoles and electron dense granules, nuclear fragmentation and necrosis. In Cur groups,the morphological changes of apoptosis above mentioned were also found and the cellular oncosis,which would result in necrosis with karyorhexis and cell swelling,were observed. Conclusion Ele and Cur can induce the apoptosis and/or oncosis of LECs. This result provides a scientific basis for pursuing safe and effective natural drugs for preventing and treating after cataract.
