摘 要:目的:探讨大鼠自身血栓微颗粒诱导其心脏心内膜下微血管内血栓形成的机制。方法:通过向大鼠主动脉根部注入自身血栓微颗粒建立冠状动脉微血栓模型,于术后1h、1天、1周、2周、3周和4周取标本。HE染色观察心内膜下微血管内血栓形成情况;酶联免疫吸附双抗体夹心法(ELISA)检测血清中vonWillebrand因子(vWF)含量。结果:HE染色结果显示心内膜下微血管内有血栓形成,部分血栓与内膜紧密粘连。向大鼠体内注入自身血栓微颗粒后1h,血浆中vWF水平显著增高。结论:大鼠自身血栓微颗粒能够诱导其冠状动脉微血栓形成,其心脏心内膜下微血管内皮细胞活化是可能参与机制之一。Objective: To investigate the mechanism of rat endocardial microvascular thrombosis induced by microthrombotic particles. Method: The model of coronary artery microthrombosis was constructed by aortic root injection with microthrombotic particles. The sample was taken 1h, 1d,1w, 2w, 3w and 4w after operation respectively, dyed through HE coloration.Endocardial microvascular thrombosis was observed by the optic microscope. The levels of von Willebrand Factor (vWF) in sera were detected using enzyme-linked immunosorbent assay (ELISA).Results: Platelet thrombosis could be observed in rat endocardial microvessel,partly conglutinated tightly with vascular endothelial cells. The vWF levels increased remarkably 1h after operation. Conclusion: It is suggested that the microthrombotic particles induce rat coronary artery microvascular thrombosis through the activation of endocardial microvascular endothelial cells.
