摘 要:目的:探讨放射损伤小鼠骨髓组织黏附分子ICAM1的表达和基质细胞黏附能力的变化及川芎嗪对其影响。方法:健康昆明小鼠经6.0Gy60Coγ照射后立即喂饲川芎嗪并设对照组和正常组,在放射损伤后第3、7、14和21天检测其骨髓有核细胞(BMMNC)和骨髓成纤维细胞集落形成单位(CFUF),骨髓黏附分子ICAM1的表达水平以及骨髓基质细胞黏附能力。结果:川芎嗪能够减轻放射损伤小鼠骨髓有核细胞、骨髓成纤维细胞集落形成单位(CFUF)和骨髓基质细胞黏附能力的下降。照射后第3、7天骨髓黏附分子ICAM1的表达水平显著增高,并且川芎嗪组ICAM1表达水平高于对照组;但照射后第14天川芎嗪组ICAM1表达水平开始下降;而照射组ICAM1表达水平仍持续增高,照射后第21天川芎嗪组ICAM1表达水平恢复正常(P>0.05),而对照组才开始下降,二者比较有显著差异(P<0.01)。结论:急性放射损伤早期骨髓黏附分子ICAM1呈高表达。川芎嗪促进放射损伤小鼠骨髓造血恢复的机制可能是通过改变黏附分子的表达并进而影响黏附功能实现的。Objective:To explore the expression of adhesion molecule ICAM-1 and the adhesive capability of Stroma cells in irradiation injured mice and the significance of ligustrazine on it. Methods: Kunming strain mice were given Ligustrazine in ligustrazine group after irradiation by 6. 0 Gy^60Co γ-ray and compared with control group and normal group. On the 3rd,7th, 14th and 21st day after irradiation, the bone marrow mononuclear cells( BMMNC), and the fibroblast colony forming units(CFU-F) were counted; the exprestion level of adhesion molecule ICAM-1 as well as the adhesive capability of Stroma cells was evaluated too. Results: Ligustrazine can mitigate the drop of the counts of BMMNC, CFU-F, and he adhesive capability of Stroma cells. On the 3rd and 7th day after irradiation, the expression of adhesion molecule ICAM-1 was heightened. And it was higher in ligustrazine group than in control group. But on the 14th day after irradiation, the expression of ICAM-1 in ligustrazine group was declined and the expression level of ICAM-1 in control group was continuously heightened. On the 21st day after irradiation, the expression of ICAM-1 in 1 igustrazine group has recovered and in control group was dropping, both compare markedly difference (P 〈 0.01 ). Conclusion:The expression of adhesion molecule ICAM-1 was high in the early phase after acute irradiation. The mechanism of ligustrazine promoting hematopoiesis is likely to change the expression of adhesion molecule ICAM-1 and influence the adhesive capability of Stroma cells.
