摘  要:目的:探讨血型物质与载体固相化及其对人血浆中特异性IgG的吸收。为母婴血型不合的自然流产或死胎的早期治疗奠定实验基础。方法:用加热煮沸法分离人血型物质,用sephedex200纯化后使其与载体固相化,用无水酒精固定、生理盐水洗涤后制成免疫吸附物。用卡斯法和免疫浊度法检测吸收前后IgG抗体效价及蛋白质分子各组分含量。结果:高效价孕妇血浆经免疫吸附物吸收5分钟后,IgG平均由627.2降至35(P<0.05);10分钟降至19.4,IgM可完全吸收,12小时后降至11(P<0.05),更换吸附物经5分钟再次吸收,IgG可完全吸收。而血浆白蛋白、球蛋白和补体成分经不同的时间吸收后,其含量均无明显差异(P>0.05)。结论:血型物质经固相化后对特异性IgG有很强的吸收作用,可为母婴血型不合的自然流产或死胎者开辟治疗新途径。Objective:To investigate the effect of blood type substance solidified with vector on the specific absorption of the plasma IgG antibody in order to provide the experimental basis for the early treatment of spontaneous abortion or stillbirth due to fetomateral ABO blood typing incompatibility. Methods :The human blood type substance was isolated via heating and purified by sephedex-200, favoring its solidification with the vector. Then it was stabilized by 99. 8% Alcohol and washed in 0. 9% injective saline, resulting in the formation of immune absorbed substance. The titer of IgG antibody was assayed by kars method and the protein concentration was determined by immnue turbidity method before and after the absorption. Results:5 min,10 min and 12 h after absorption with immune absorbed substance, the high titer of plasma IgG in the pregnant women was, on average, reduced trom 627.2 to 53, 19. 4 and 11 (P 〈 0. 05), respectively. Whereas IgM was completely absorbed. And IgG could be also absorbed totally 5 rain after the immune absorbed substance was replaced with the fresh one. During the whole process there was no significant change in the level of plasma albumin globulin or complement(P 〉 0. 05). Conclusion:The blood type substance solidified with the vector has a great potency in absorbing the specific IgG antibody, which can provide a new approach in the management of the sporitaneous abortion or stillbirth caused by fetomateral ABO blood typing incompatibility.
