摘  要:目的探讨丹参酮IIA对人肝癌细胞TGF-β1表达的影响及其抗肿瘤作用机制。方法体外培养SMMC-7721肝癌细胞株,采用倒置相差显微镜、Hoechst33342染色荧光显微镜观察,以及“梯状”DNA确定凋亡的发生并计算凋亡率;用RT-PCR方法检测肝癌细胞TGF-β1的表达。结果倒置相差显微镜观察可见各试验组细胞的生长不同程度受抑,并可见典型的凋亡形态改变;Hoechst33342染色可见凋亡细胞核浓染,亮度明显加深或有染色质边集现象,亦可见典型的凋亡小体;提取基因组DNA电泳检测,出现典型的梯状条带;RT-PCR检测显示,丹参酮IIA作用48小时后TGF-β1表达量随药物浓度的增大而逐渐减低。结论丹参酮IIA可抑制人肝癌细胞SMMC-7721的细胞增殖,促进其凋亡,此作用可能与细胞内TGF-β1表达下调有关。Objective To evaluate the effects of Tanshinone ⅡA on SMMC-7721 apoptosis and the change of gene expression of TGF-β1, and investigate the possible antitumor mechanism of Tanshinone ⅡA. Methods We observed the phenomenon of apoptosis with the ,inverted phase contrast microscope, by staining with Hoechst33342 and the DNA ladder; RTPCR was used to detect the gene expression of TGF-β1. Results Inhibition of cell proliferation and apoptosis was observed with the inverted phase contrast microscope ; Staining with Hoechst33342 and the DNA ladder manifested the existence of apoptosis ; The gene expression of TGF-β1 was downregulated while the concentration of Tanshinone HA was increasing. Conclusion Tanshinone ⅡA can inhibit cell proliferation and induce apoptosis in vitro, and the results maybe relates to the downregulation of gene expression of TGF-β1.
