高效液相色谱法测定康力宝口服液中麦角甾苷含量

摘　　要：目的 建立康力宝口服液中麦角甾苷的高效液相色谱含量测定方法。方法 采用Shim-pack vp-ODS柱（4．6mm×150mm，5μm），以甲醇-乙腈-四氢呋喃-0．1mol·L^-1醋酸钠缓冲液（冰醋酸调pH至4．0）（8：8：7：77）为流动相，流速为1．0ml／min，检测波长为334nm，灵敏度为0．05AUFS。结果 麦角甾苷在4．05～64．80μg·ml^-1范围内呈良好的线性关系，r=0．9998。平均回收率99．65％，RSD为2．10％（n=5）。结论 方法简便、准确、重现性好，可用于该制剂的质量控制。Aim To establish a method for the determination of acteoside in Kanglibao oral liquid by high performance liquid chromatography （HPLC）. Methods The chromatographic column ： Shim - pack vp - ODS column. Mobile phase ： methanol - acetonitrile - tetrahydrofuran - 0.1 mol · L^- 1 sodium acetate buffer （ adjust pH to 4.0 with Acetic acid glacial） （ 8： 8： 7：77 ）. Flow rate ： 1.0ml/min. Detection wavelength ：334nm. Sensitibity ：0.05AUFS. Results The acteoside curve was linear in the range of 4.05 - 64.80μg · ml^ - 1 （ r = 0.999 8 ）. The average recovery of the method was 99.65%, RSD = 2.10% （ n = 5 ）. Conclusions The method was simple, accurate and reproducible and could be used for the quality control of this preparation.
