补阳还五汤及黄芪体外对家兔血小板活化因子受体活性的影响

摘　　要：目的:探讨补阳还五汤及黄芪体外对家兔血小板活化因子(PAF)受体活性的作用,阐明黄芪在该方中的配伍机制。方法:将动物分为补阳还五汤全方组、活血组和补气组,采用放射配基受体结合实验,以家兔血小板PAF受体特异结合抑制率为评价指标,探讨3组体外对家兔血小板PAF受体活性的影响。结果:3组均能抑制3H-PAF与家兔血小板PAF受体的特异结合,对PAF受体特异结合抑制程度由大到小依次为活血组、全方组、补气组,各组之间抑制率差异均有显著性(P<0.01)。结论:单味黄芪(120g)对家兔血小板PAF受体与3H-PAF的特异结合有一定的抑制作用,但黄芪与方中活血药配伍后,能使后者拮抗血小板PAF受体的作用明显减弱,使全方拮抗血小板PAF受体的作用缓和,防止大量活血药攻伐太过,体现了补阳还五汤“祛瘀而不伤正”的配伍机制。Objective：To study the effects of Buyang Huanwu Decoction（BHD） and Astragalus mongholicus on in vitro platelet activating factor （PAF） receptor activity in the platelet of the rabbit. Methods .. The study included three groups,whole BHD group ,group of activating blood circulation and group of invigorating qi,and the specific binding inhibition rate of PAF receptor was determined with radioligand receptor binding method. Effects on in vitro PAF receptor activity in the platelet of the rabbit were compared among the three groups, Results：The specific binding of ^3H-PAF with PAF receptor in the platelet of the rabbit was inhibited in all the three groups ,and the inhibiting degree was in the following order ;group of activating blood circulation〉 whole BHD group 〉 group of invigorating qi with significant differences among the three groups （P〈 0. 01）. Conclusion： Single Astragalus mongholicus（120g） can inhibit the specific binding of PAF receptor in the platelet of the rabbit with ^3H-PAF ,but combination of Astragalus mongholicus with the drugs for activating blood circulation in BHD can significantly decrease the inhibiting action of the latter on PAF receptor activity of the platelet ,reflecting the combined mechanism of ＂ removing blood stasis without injuring the votal qi＂ in BHD.
