摘  要:目的利用C57BL/6小鼠制备H-Y抗血清并分析。方法以纯系C57BL/6雄鼠脾细胞做为抗原,通过腹腔注射免疫雌鼠的方法制备H-Y抗血清。利用经典精子细胞毒实验和ELISA检测方法分别对两组抗血清检测。结果H-Y抗血清对鼠精子的平均工作效价为1/20—1/160,而ELISA检测抗血清的最高效价为11640,比精子细胞毒实验的检测结果高。结论对两种检测方法的结果进行分析,认为ELISA能准确判断出H-Y抗血清的特异性和效价,尤其在选择免疫效果较好的雌鼠方面有较高的参考价值。Objective To prepare and analyze H-Y antiserum from C57BL/6 mice. Methods Isogenic C57BL/6 female mice were immunized with male spleen cells as a serum donor in the search for specific antibodies against H-Y antigen. To separate immunized female mice showing higher titer antiserum. The antibody in sera of the two groups of immunized female mice were assayed by sperm cytotoxicity test or ELISA technique. Results Sperm cytotoxicity test gave a H-Y polyclonal antibody titer of 160. ELISA analysis showed that H-Y polyclonal antibody had a significant binding activity with male specific antigen, possessing a serum antibody titer of 1 : 640. Conclusion In comparison of results of the two methods there is a significant difference in antibody titers between those two methods. The specificity and titer of the H-Y antisera can be batter identified by ELISA technique.
