无/低血清条件下杂交瘤细胞系生长和McAb产生的研究

摘　　要：将3G1/3F8细胞培养于血清浓度为20％、10％、17.5％、5％、2％、1％、0％的RPMI-1640中测定细胞生长曲线和McAb产生曲线。观察血清浓度对细胞生长和McAb产生的影响。用6种无血清培养基对3G1/3F8细胞做适应性培养,活细胞数为对照组的9.5％～55％;McAb产量为81.8％～106.8％。经驯化培养3G1/3F8细胞能够在血清含量1％、5％的培养液生长传代,冻存复苏,活细胞数达到对照组的71.2％～98.2％;McAb产量为104％和108％。对不同血清浓度下接种密度对细胞生长和McAb产量的作用做了实验观察。Hybridoma cell line (3G1/3F8) was cultivated in RPMI-1640 medium with different serum concentration (20%, 10%, 7.3%, 5%, 1 %, 0%,) to observe effect of serum concentration on cell growth and Mc/Vb production. The cel.t line was cultivated in six serum free medium respectively for adaption. Viable cells number obtained from serum -free medium was 9.5%-55% of that from serum medium.McAb concentration was 81.8%-106. 8%. The cell line could grow, propagate, freeze and recove in RPMI-1640 with 1% and 5% serum by domesticating. Viable cells number obtained from low serum medium was 72. 1 %-98.2% of that from high serum medium. McAd , concentration was 104%-108%. The cell was cutivated RPMI-1640 medium with different concentration at varius initial cell density. Cell growth and McAb production were observed
