摘  要:目的探讨一种制备小口径异种移植血管的新方法,为冠状动脉旁路移植术提供新的移植血管来源。方法利用酶-去污剂法去除犬颈动脉血管壁结构细胞,脱细胞后其中一部分再进行肝素结合,组织学及机械性能研究观察脱细胞效果。24只兔的左、右两侧颈动脉同时分别植入结合与未结合肝素的脱细胞异种血管,作为自身对照,分别于3周、3月和6月,用B超观察移植血管的通畅情况。术后3月随机处死12只兔,取材后行Ⅷ因子和α-肌动蛋白免疫组化检查。结果脱细胞后管壁无细胞成分残留,而细胞外基质保存完好。植入兔颈动脉后两组均无血管阻塞,而肝素结合组血栓形成少于未结合组。3月后管壁出现平滑肌细胞,而内腔由内皮细胞覆盖。结论酶-去污剂联合肝素结合处理是制备小口径异种移植血管可行的方法。Objective To explore a new small-caliber vascular xenograft for coronary artery bypass grafting. Methods Canine carotid arteries were decellularized by detergent and enzyme. A part of decellularized vascular gratis were covalently linked with heparin. Histological examination and mechanical test were performed. Twenty-four rabbits were performed bilateral carotid bypass grafting. Heparin-linked and nonheparin-linked xenografts were implanted in left and right carotid artery respectively. Graft patency was checked by uhrasonography at 3 weeks, 3 and 6 months. Twelve rabbits were randomly killed and gratis were harvested at 3 months. Immunohistochemical staining for Factor Ⅷ and α-actin were performed. Results All the cells of these canine carotid arteries were removed with the extracellular matrix well observed. There was no obstruction at both sides after implantation, while less thrombus was found in the decellularized heparin-linked grafts than in the contralateral side. Smooth muscle cells densely populated the graft wall and endothelial cells covered the vascular lumen of xenograft at 3 months. Conclusion A new small-caliber vascular xenograft may be effectively developed through detergent and enzyme and heparin linkage.
