摘  要:目的:观察2型糖尿病患者硝化酪氨酸、谷胱甘肽过氧化物酶水平以及与年龄、性别、体质量指数、血压、糖代谢、脂代谢等指标的关系。方法:于2004-10/2005-03选择哈尔滨医科大学第一临床医院内分泌科收治的2型糖尿病患者55例为研究对象,选择同期自愿参加调查研究的健康体检者50名作为对照。采用葡萄糖氧化酶法测定空腹血糖及餐后2h血糖,采用高压液相离子交换层析法测定糖化血红蛋白,采用全自动生化分析仪测定总胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇。采用酶联免疫吸附法检测硝化酪氨酸,采用比色法检测谷胱甘肽过氧化物酶。分析硝化酪氨酸及谷胱甘肽过氧化物酶与年龄、性别、体质量指数、血压、糖代谢、脂代谢等指标的相关性。结果:2型糖尿病组55例和正常对照组50名均进入结果分析。①2型糖尿病组硝化酪氨酸水平明显高于正常对照组[(98.6±15.2),(59.0±8.7)nmol/L,P<0.01]。②2型糖尿病组谷胱甘肽过氧化物酶活性低于正常对照组[(0.8±0.1),(1.2±0.1)mkat/L,P<0.05]。③为平衡年龄、性别、体质量指数、腰臀比、血压、空腹血糖、糖化血红蛋白、三酰甘油对硝化酪氨酸及谷胱甘肽过氧化物酶的影响,以年龄、性别、体质量指数、收缩压等因素为协变量,分析显示2型糖尿病组硝化酪氨酸水平显著高于正常对照组(P<0.01),2型糖尿病组谷胱甘肽过氧化物酶活性显著低于正常对照组(P<0.05)。硝化酪氨酸与空腹血糖正相关(r=0.63,P<0.01),谷胱甘肽过氧化物酶活性与糖化血红蛋白负相关(r=-0.54,P<0.01)。血清硝化酪氨酸水平及谷胱甘肽过氧化物酶活性与年龄、性别、体质量指数、收缩压、舒张压、餐后2h血糖、胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇无相关性。结论:糖尿病患者硝化酪氨酸水平升高,谷胱甘肽过氧化物�AIM: To observe the levels Of nitrotyrosine and glutathione peroxidase and their relationship with age, sex, body mass index, blood pressure, glycometabolism and lipometabolism in patients with type 2 diabetes. METHODS: Totally 55 outpatients with type 2 diabetes mellitus, who were treated in Department of Endocr/nology, First Clinical Hospital, Harbin Medical University between October 2004 and March 2005, were enrolled. Another 50 health volunteers were taken as the normal controls at the same time. Fasting glucose and blood glucose after 2-hour meal were measured using the glucose oxidase method. Glycosylated hemoglobin was determined with high-pressure liquid ion exchange chromatography. Total cholesterol, triglycerides, high density lipoprotein cholesterol and low density lipoprotein cholesterol were measured via the full automatic biochemical analysor. The enzyme linked immunosorbent assay and chromatometry were used to determine the nitrotyrosine and glutathione peroxidase. Correlation of nitrotyrosine and glutathione peroxidase with age, sex, body mass index, blood pressure, glycometabolism and lipometabolism was analyzed. RESULTS: Totally 55 patients of type 2 diabetes and 50 healthy testees were involved in the result analysis. ①Level of nitrotyrosine was markedly higher in patients with diabetes than in control subjects [(98.6±15.2); (59.0±8.7) nmol/L ;P 〈 0.01 ]. ②Glutathione peroxidase was decreased in the diabetes group as compared with the normal control group [(0.8±0.1), (1.2±0.1) mkat/L,P 〈 0.05]. ③With mean age, sex, body mass index, waist-to-hip ratio, blood pressure, fasting plasma glucose, glycosylated hemoglobin and triacylglycero as covar/ate, and age, sex, body mass index, systolic blood pressure and so on as concomitant variable, the level of nitrotyrosine was obviously higher in patients with diabetes as compared with the normal control group(P 〈 0.01 ).
