摘  要:目的:观察核心结合因子α1在SD大鼠胫骨骨折愈合过程中表达的变化,探讨核心结合因子α1在骨折修复中的作用。方法:实验于2003-10/2005-02在教育部口腔生物医学工程重点实验室完成。①将48只6个月龄雌性SD大鼠左侧胫骨制成相同的骨折模型,采用克氏针行髓腔内固定。②于骨折术后7,14,21,28d分批处死动物,采用X射线、组织学及免疫组织化学、半定量反转录-聚合酶链反应方法观察核心结合因子α1在骨折愈合过程中不同时期表达情况。结果:①X射线显示胫骨骨折断端无移位。②组织学切片显示骨折愈合过程中可同时见纤维性骨痂和软骨性骨痂形成。③核心结合因子α1免疫组织化学染色显示骨折术后第7天及14天时在增殖的间充质细胞及软骨细胞中见强阳性反应,从第21天起,核心结合因子α1染色阳性细胞数及阳性反应程度明显下降,7,14,21,28d各时间点免疫组织化学染色的平均吸光度值分别为0.568±0.013,0.534±0.016,0.313±0.011,0.277±0.008。④反转录-聚合酶链反应结果表明骨折术后第7天及14天时核心结合因子α1mRNA表达量较高,第21天及28天时,核心结合因子α1mRNA表达下调,各时间点基因表达的相对强度均值分别为1.935±0.231,2.103±0.264,0.956±0.045,0.527±0.038。结论:在SD大鼠胫骨骨折髓腔内内固定模型中,骨愈合主要表现为二期愈合方式,核心结合因子α1集中表达在骨痂形成的早期,说明核心结合因子α1是骨折愈合的启动调节因子之一。AIM: To observe the change of the expression of core binding factor (CBF) α1 in the healing processes of tibia fracture in SD rats so as to evaluate the function of CBF α1 on fracture healing. METHODS: The experiment was completed at the Key Lab for Oral Biomedical Engineering of Ministry of Education from October 2003 to February 2005. ①Standardized fracture models of left tibia in 6 months old female SD rats (n=48) were induced, and intramedullary stabilization was performed with Kirschner-wires. ②At days 7, 14, 21 and 28 after fracture the animals were killed in batch. The expression of CBF α1 in different fracture healing phases was observed with X-ray, histology, immunohistochemistry, semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: ①In tibia fracture healing, no dislocation was occurred in X-ray; ②provisional callus and endochondral callus were observed in tibia fracture healing under histological sections; ③ Immunohistochemical staining with intense positive reactions were detected in mesenchymal cells and chondrocytes in the periosteum and marrow near the fracture site at day 7 or day 14 after the operation. 21 days later, the cells expressing the CBF α1 declined obviously. The mean absorbance (A) measured by immunohistochemical staining was 0.568±0.013,0.534±0.016,0.313±0.011, 0.277±0.008, respectively at days 7, 14, 21 and 28 after the operation; ④ The level of CBF α1 gene mRNA was higher at days 7 and 14 by RT-PCR reaction; the expression of CBF α1 gene mRNA was down-regulation at days 21 and 28. The mean relative density was 1.935±0.231,2.103±0.264, 0.956±0.045,0.527±0.038, respectively at days 7, 14, 21 and 28 after the operation. CONCLUSION: The expression of CBF α1 takes place in early phase of bone callus formation during the processes of secondary fracture healing in the internal fixation models of SD rats. It is indicated that the CBF α1 is one of the promoting regulatory factors
