摘 要:目的:观察福辛普利晚期预处理对缺氧复氧心室肌细胞L型钙通道电流的影响。方法:应用全细胞膜片钳方法,记录酶解的成年豚鼠心室肌细胞及福辛普利预处理后L型钙通道电流在缺氧复氧期的变化。结果:缺氧复氧期心室肌细胞L钙电流峰值明显下降,与对照组相比有明显差异(-237.71±17.70pAvs-286.00±21.15pA,P=0.0006,n=7),L型钙电流电流-电压曲线上移;福辛普利预处理后心室肌细胞L钙电流峰值亦下降,但下降不明显(-265.57±20.20PAvs-286.00±21.15pA,P=0.089,n=7)与缺氧复氧组比较有明显差异(P=0.018),L型钙电流电流-电压曲线上移幅度减小。结论:福辛普利可稳定缺氧复氧心肌L型钙通道电流,对心肌有保护作用。Objective: To investigate effect on L type calcium currents of fosinopril late preconditioning in ventricular myocytes during anoxia/reoxygenation period. Methods: Using whole-patch clamp cinefiguration in isolated adult guinea pig ventricular myocytes with an enzymatic procedure the L-type calcium current were measured before and after ischemic application of each group respectively. Results: Under the condition of reoxygenation, the peak of ICa-L reduced significantly(-237.71±17.70 pA vs. -286.00±21.15 pA,P=0. 0006,n= 7) and ICa-L Ⅰ~Ⅴ curve shifted to upper. After fosinopril preconditioning, the peak of ICa-L insignificantly reduced compared to control (-265.57± 20. 20 PA vs. ±286.00± 21.15 pA, P= 0. 089, n= 7), but compared with reperfusion group its difference to reperfusion group was significant (P=0. 018), its ICa-L Ⅰ~Ⅴ curve shifted to under. Conclusion: Fosinopril can stabilize L type calcium currents in myocytes during anoxia/reoxygenation, so possess the protective effect for ventricular myocytes.
