摘 要:[目的]观察葡萄糖酸锌、亚硒酸钠单独及联合作用对二氧化硅致成纤维细胞DNA损伤的影响,寻找两者的最佳剂量组合。[方法]生长良好的中国仓鼠肺成纤维细胞,以5×108个/L的密度接种到25ml培养瓶中,经24h培养后加入终浓度为100mg/L的SiO2,同时分别加入终浓度分别为1.0、1.5、2.25mg/L的葡萄糖酸锌(ZnG)和终浓度分别为0.5、1.0、2.0μmol/L的亚硒酸钠(Na2SeO3)及各浓度的ZnG+Na2SeO3交互组合。作用2h后收获细胞,调细胞密度为1×109个/L,通过彗星试验分别观察DNA的损伤情况。同时设阴性对照组(生理盐水)、阳性对照组(终浓度为100mg/L的SiO2)、ZnG对照组(2.25mg/L)、Na2SeO3对照组(2.0μmol/L)。联合作用时,采用L(34)正交设计表进行试验。9[结果]与SiO2组比较,1.0、1.5、2.25mg/L的ZnG及0.5、1.0、2.0μmol/L的Na2SeO3均可以显著减轻SiO2致成纤维细胞DNA链的断裂作用(P<0.05);2.25mg/L的ZnG与0.5μmol/L的Na2SeO3联合作用效果最好。[结论]ZnG、Na2SeO3单独和联合作用在体外可有效减轻SiO2粉尘所致成纤维细胞DNA的损伤作用。[ Objective ] To investigate the effect of Zinc gluconate and sodium selenite which were used separately and combined on DNA damage in Chinese hamster lung cells treated with silicon dioxide. [ Methods ] Chinese hamster lung cells were cultured with silica( terminal eoncentration 100 mg/L ), at the same time Zinc gluconate( terminal concentration 1.0, 1.5,2.25 mg/L ) and Sodium selenite ( terminal concentration 0.5, 1.0, 2.0 μmol/L ) were added separately and combined respectively. DNA damage were measured through comet assay. [ Results ] Zinc gluconate( 1.0, 1.5,2.25 mg/L ), sodium selenite( 0.5, 1.0, 2.0 μmol/L )and Zinc gluconate combined with sodium selenite have the effects to reduce DNA damage in Chinese hamster lung cells treated with silica ( P 〈 0.05 ). The most suitable combination of the two compounds should he the highest dosage of Zinc gluconate ( 2.25 mg/L )with the lowest dosage of sodium selenite ( 0.5 μmol/L )( P 〈 0.05 ). [ Conclusion ] Zinc gluconate and sodium selenite which were used separately and/or combined could reduce DNA damage in Chinese hamster lung cells exposed to silica in vitro.
