Effect of subchronic exposure to 2,3,7,8-tetrachlorodibenzop-dioxin(TCDD) on reproductive system of male rats

摘  要:目的了解2,3,7,8-四氯二苯并二噁英(TCDD)对雄性大鼠生殖系统的影响。方法清洁级Wistar雄性大鼠32只,体重(100±10)g,随机分成4组,每组8只,连续经口灌胃染毒90 d,剂量分别为2.5,25,250 ng/kg,对照组给予二甲基亚砜(DMSO),测定血清中睾酮(T)、促黄体生成素(LH)、促卵泡生成素(FSH)的激素水平以及睾丸、前列腺、精囊腺的脏器质量系数,附睾尾精子畸形率。结果与对照组比较,各染毒组T水平下降,差异有显著性(P<0.05);各染毒组FSH和LH水平上升,但差异无显著性(P>0.05);中、高剂量组的睾丸、精囊腺及所有染毒组的前列腺脏器质量系数均低于对照组(P<0.05);各染毒组精子畸形率随剂量的增加而上升,中、高剂量组与对照组相比差异显著(P<0.01)。结论TCDD亚慢性染毒,可影响精子和生殖系统的正常发育,并在一定程度上对生殖激素的稳态造成了干扰。Objective To study the effect of TCDD on reproductive system of male rat. Method Thirty two healthy male Wistar rats were randomly divided into four groups according to body weight ( 100 ± 10) g: control group (rats were orally given 250 ng/kg dimethyl sulfoxide, DMSO), and three exposed groups (orally received different doses of TCDD-2.5, 25 and 250 ng/kg per day, respectively for 90 days). At the end of experimental period, the animals were fasted overnight, then weighed and sacrificed. Testicle, epididymis, prostate and seminal vesicle were removed for measuring their relative weights to body weight. Meanwhile, the serum levels of testrone (T), luteinizing hormone (LH), follicle-stimulating hormone (FSH) and the sperm abnormality rate in epididymis tail were also measured. Result The results showed that the sentm testosterone levels and weight coefficients of prostate in rats of all the TCDD exposed groups were significantly lower than that of controls ( P 〈 0.05), the serum levels of FSH and LH also seemed to show some increase but without any significant difference compared with controls ( P 〉 0.05). Additionally, the weight coefficients of testicle, epididymis and seminal vesicle in middle or high TCDD exposed groups, and the percentages of sperm abnormalities in epididymis tail of rats in middle or high TCDD exposed groups were significantly increased compared with controls ( P 〈 0.05). Conclusion It is showed that subchronic exposure to TCDD could damage the normal development of sperm and reproductive system, and disturb the homeostasis of reproductive hormones suggesting that dioxins might affact the function of reproductive-endocrine system in male rats to a certain extent.
